Spheroides rubripes is most commonly found along the sea coasts of China and it may also be found in Korea, Japan and Vietnam. Spheroides rubripes liver oil has been merely used for making soap and other industrial purposes, because it contains a commonly known toxin, called tetrodoxin. However, the tetrodoxin is completely soluble in water and is readily soluble in hot water...

Spheroides rubripes liver oil has been merely used for making soap and other industrial purposes, because it contains a commonly known toxin, called tetrodoxin. However, the tetrodoxin is completely soluble in water and is readily soluble in hot water and is hydrolized in alkaline solutions. Thus the oil can be prepared free from tetradoxin. In this study, the oil is proved to be nontoxic by a series of acute and subacute toxicity tests in mice and rats. Pharmacological studies of the oil in dogs, mice, rats, and frogs have been made and it is proved that there is no untoward symptoms and signs which indicate toxic reactions.

红鳍圆鲀(Spheroides rubripes)盛产于我国沿海一带,其肝含油量高达52—67.05%,维生素A含量也较丰富(530—2535国际单位/克),但因其肝脏含有河豚毒素,过去其油仅作照明、制造肥皂等工业用. 根据河豚毒素易溶于水,不溶于脂肪油,更易溶于热水,遇碱易破坏的特性制成甲种和乙种红鳍圆鲀鱼肝油,通过小白鼠、大白鼠的急性、亚急性毒性观察和对狗、小白鼠、大白鼠、青蛙的一般药理实验证明如此制成的鱼肝油无明显的毒性反应。

A systematic approach to evaluate the effects of phenylalanine and phenylacetate on developing nervous tissue is reported. Choline acetyltransferase(CAT) was decreased in those cultures exposed to phenylalanine(0.6mM) by 40%, and to phenylacetate(0.6mM) by 26—38%.These preliminary data suggest that both phenylalanine and phenylacetate are detrimental to certain growth aspects of immature mouse cortical neurons; furthermore, phenylacetate appears to be more toxic than phenylalanine at similar concentrations.

我们以16天孕龄的胎鼠大脑皮层原代介离性体外培养方法为基础,观察0.6毫克分子浓度的苯丙氨酸与苯乙酸各自对发育中脑神经元的影响。结果显示:在苯丙氨酸及苯乙酸加入培养液中14—18天后,产生培养中神经细胞的破伤风毒素特殊结合量及3H—GABA特殊摄入量减少;胆碱乙酰移换酶及谷氨酸脱羧酶活力下降。推测以上改变乃由于神经元而非胶质细胞的数量有所减少,苯乙酸对于未成熟脑神经细胞的毒性似较苯丙氨酸强烈。

In previous studies,we found that toosendanin,a triterpenoid derivative ofplant origin had a selective and irreversible blocking action on presynaptic terminalsat neuromuscular junction similar to that of botulinum toxin and β-bungarotoxin.In this work,to search for the binding site of toosendanin,the large synaptosomefraction was isolated from the cerebral cortex of rat and was incubated with too-sendanin in the physiological medium(pH,temperature and ionic strength),andthen the concentration of toosendanin in the supernatant was determined.The results obtained showed that the concentration of toosendanin had signi-ficantly decreased in the supernatant after it was incubated with synaptosome frac-tion.The decrease was also related with the incubating time,temperature andionic strength in the medium.It diminished as the incubating temperature fellbelow 20℃ or the incubating time was less than 20 min;the largest decrease wasobserved at an ionic strength of 0.15.Only the grey substance,not the white substan-ce or liver,was able to decrease the concentration of toosendanin in the supernatantwhen they were incubated with toosendanin.These results indicated that itwas the synaptosome in the nerve system that selectively bound toosendanin.

将大鼠大脑皮层勻浆经离心分离的大突触体组份同含一定量的川棟素溶液于生理条件(温度、pH、离子强度)下保温,再经离心分离后,用对二甲氨基苯甲醛试剂显色的分光光度法测定上清液中的川棟素含量,结果表明上清液中川棟素含量明显降低,在一定范围内,降低的程度随大突触体组分浓度的提高而增加。而同样处理的肝组织、脑白质与川棟素保温后,上清液中川棟素含量基本不变。由这些结果看来,可能川棟素同突触体有一选择性的结合。
