An optimal protoplast yield of up to 90% was obtained in 2-3 hours by treating 5-6 day-old leavea with an enzyme mixture of cellulase EA3-867 (2%) and pectinase (0.5%) in 0.65 M mannitol at 24-26℃. Protoplasts from mesophyll cells of rye plants (Secale cereale L.) were isolated by enzymatic removal of cell walls in an osmoticum consisting of mannitol. An optimal protoplast yield of up to 90% was obtained in 2-3 hours by treating 5-6 day-old leavea with an enzyme mixture of cellulase EA3-867 (2%) and pectinase (0.5%) in 0.65 M mannitol at 24-26℃. The protoplasts cultured at a density of 5 104 to 105/ml in a liquid medium (table 1) supplemented with 2, 4-D (1 mg/l), 6 BA (0.5 mg/l), CH (1 g/l) and 0.56 M sucrose, and placed in a controlled growth cabinet maintained at 23-26℃, and illuminated with fluorescent light of about 800-2000 lux. They were able to regenerate cell walls within 48 hours. They underwent first division in 3-7 days and formed cell clusters in 9 days. Large cell clusters were formed within 25 days of culturing. After 2 months in liquid culture medium, the microcalli were produced which were visible to the naked eye. This culture medium contained the same nutrients as the protoplasts culture media, except that the concentration of sucrose was lowered to 0.4 M.

黑麦叶肉原生质体系用纤维素酶溶去壁后分离出来的。酶液混合液为:纤维素酶(2%)、果胶酶(0.5%)溶于0.65M的甘露醇中,保温在24~26℃。将生长5—6天的黑麦叶片在酶掖中处理2—3小时后,就可使90%以上的原生质体分离下来。将每毫升含有5×10~4—10~5密度的原生质体培养在液体培养基中(表1),其中附加2,4-D(1毫克/升)、6BA(0.5毫克/升)、CH(1克/升)和0.56M蔗糖。培养条件为光照800—2000米烛光,温度23—26℃。培养48小时后长出新壁。第3天出现第一次分裂,4—7天进行第二、三次分裂,9天后形成细胞团,25天后长成大细咆团。此后,定期加等体积的新鲜培养液,继续培养一个时期后便形成了幼小愈伤组织。
