Objective: To study the existence of methylation of p16 gene in promoter region in children with nephroblastoma and the relation between methylation and histologic type of the tumor as well as the prognosis. Methods: Twenty cases with nephroblastoma were observed on by 3 kinds of enzyme (Fnu D Ⅱ, Sac Ⅱ, Hpa Ⅱ), and 5 normal kidneys served as control. 目的:观察小儿肾母细胞瘤p16基因启动子区有无甲基化及有甲基化者与组织类型和预后之间关系。方法:20例肾母细胞瘤(5例肾组织作对照)进行甲基化观察,使用三种内切酶分别为FnuDⅡ、SacⅡ、HpaⅡ。结果:肿瘤组有6例出现甲基化(对照组无甲基化),除1例属于预后良好型(FH)之外,余5例均属预后不良型(UH)。甲基化与组织类型有关。随访结果有甲基化病例(UH组),1例术后1.5年死亡,1例术后半年复发,证明甲基化影响着预后。结论:肾母细胞瘤p16基因启动子区存在甲基化,甲基化者属于组织预后不良类型者多。

Results p16 gene deletion was found in 11 of 52(21.15%) BTCCs, only one tumor showed point mutation on p16 exon 2, ninteen cases of BTCCs (36.53%) showed p16 gene 5′CpG island methylation. Conclusion There was a statistically significant association between p16 gene deletion and early stage, well differentiated tumors. Additionally, a statistically significant correlation was also noticed between p16 gene 5′CpG island methylation and tumors in late stage with poor cell differentiation.

目的明确p16基因在膀胱移行细胞癌中的改变。方法采用Southern杂交、聚合酶链反应单链构象多态性分析(PCRSSCP)及DNA甲基化分析技术分别检测52例膀胱移行细胞癌中p16基因的缺失、突变及5′CpG岛甲基化。结果52例膀胱移行细胞癌中11例(2115%)存在p16基因的缺失;仅1例肿瘤于p16基因外显子2出现点突变;19例(36.5%)肿瘤发现p16基因5′CpG岛甲基化。结论基因的缺失和5′CpG岛甲基化是p16基因在膀胱移行细胞癌中失活的主要机制。p16基因缺失多见于早期、高分化的肿瘤,而5′CpG岛的甲基化多见于晚期、低分化的肿瘤。

目的 :探讨人前列腺癌细胞转移潜能与KAI1基因表达的关系。方法 :运用Northern印迹杂交检测KAI1基因在不同转移潜能人前列腺癌细胞中的表达 ,以甲基化分析和聚合酶链反应 单链构象多态性分析 (PCR SSCP)研究KAI1低表达的机制。结果 :在转移性人前列腺癌细胞系PC3和PC3M中不论其转移潜能强弱 ,KAI1mRNA表达均降低。甲基化分析未发现KAI1基因 5’ 启动子部位CCm5CGGG甲基化。PCR SSCP分析显示 :KAI1基因第 7外显子在PC3和PC3M均显示异常带 ,PC3M较弱。结论 :KAI1基因表达降低与人前列腺癌的转移有关 ,但其转移潜能的强弱可能不取决于KAI1的调控。KAI1基因的低表达与 5’ 启动子部位CCm5CGGG甲基化无关 ,可能与该基因的突变失活有关。
