The removal of phenol and chlorophenol from waste water catalyzed by horseradish peroxidase was studied in this paper to understand the kinetic features of this reaction.The effects of pH,H2O2 concentration and substrate type on the removal of pollutants were discussed in detail.The results showed that the optimum pH for the removal efficiency of phenol and chlorophenol was 90 in the test pH range of 50 to 90.The results also indicated that there was an optimum H2O2 concentration for pollutants...

The removal of phenol and chlorophenol from waste water catalyzed by horseradish peroxidase was studied in this paper to understand the kinetic features of this reaction.The effects of pH,H2O2 concentration and substrate type on the removal of pollutants were discussed in detail.The results showed that the optimum pH for the removal efficiency of phenol and chlorophenol was 90 in the test pH range of 50 to 90.The results also indicated that there was an optimum H2O2 concentration for pollutants removal catalyzed by horseradish peroxidase and H2O2 at concentrations higher than the optimum value had inhibition effect on pollutant removal.But the inhibition effect of higher H2O2 concentrations was very small and had little influence on the reaction.Several substrates were compared on their affinity with horseradish peroxidase according to the kinetics parameters of reactions.The results showed that phenol and 4chlorophenol were more easy to be removed than 3chlorophenol.It was also concluded in this paper that oxidation reaction catalyzed by horseradish peroxidase had the characteristic of PingPang reaction.

为了研究辣根过氧化物酶处理酚和氯酚的动力学过程,采用人工配水试验,测定了pH对酶催化反应的影响、H2O2抑制酶催化反应的动力学参数和不同底物的酶催化反应动力学参数.结果表明,在试验的pH范围内,pH为9时过氧化物酶对苯酚和对氯苯酚的去除效率最高,分别达到99.5%和95%.在较低浓度时污染物去除效率随H2O2浓度的升高而增加,较高浓度H2O2对酶催化反应有一定的抑制效应,但这种抑制作用对酶促反应的影响不大.对氯苯酚、苯酚和间氯苯酚的酶催化反应米氏常数Km依次增加,对酶的亲和性逐渐降低,间氯苯酚最难去除.根据酶反应动力学特征判定,辣根过氧化物酶催化酚形成自由基的氧化反应属于乒乓双双反应机制.

A study was carried out about the effect of high hydrostatic pressure treatment (0.1~500 MPa) under temperature (20~60℃) with dwell time 10min and pH7.0 of the enzymatic solution on horseradish peroxidase (HRP) activity and its secondary structure. It was studied by the measurement of circular dichroic(CD) spectra. The results showed that: ① The effect of compression was significant on HRP activity under the treatment temperature 40℃ and dwell time 10min and pH7.0. HRP activity increased abnormally in treatment about 100MPa, but decreaseed slowly above 400MPa. ②HRP activity decreaseed slowly under temperature of less than 40℃ at the pressure of 500MPa with dwell time 10min and pH7.0; but decreaseed rapidly with increase of temperature above 40℃. ③ There was a tight correlationship between HRP activity and the content of β-sheet conformation. The percentage rate content of β-sheet conformation decreaseed after compression, and HRP activity descended as well. Both descended rapidly after compression synergizing with mild temperature.

本文研究了超高压处理对辣根过氧化物酶活力的影响,测定了其CD谱,并分析了酶的二级结构与酶活力的关系。试验压力为0.1~500MPa,温度为20~60℃,保压时间为10min,酶溶液pH7.0。试验结果表明:(1)在处理温度为40℃、保压时间为10min和酶溶液pH7.0的条件下,压力对酶活力有显著影响;在100MPa附近的低压处理时,酶活力会反常升高;大于400MPa处理时,酶活力下降趋势缓慢。(2)在处理压力为500MPa、保压时间为10min、酶溶液pH7.0条件下,在40℃以下的温度范围内,酶的活力下降趋势缓慢;40℃以后,酶活力随温度升高下降迅速。(3)辣根过氧化物酶的活力与β-折叠的含量密切相关;超高压处理将降低酶构象中的β-折叠构象比例,从而使酶失活;温度协同超高压处理将加剧β-折叠的含量的下降,从而加速辣根过氧化物酶活力的降低。
