    The effect of aqueous ethanol (50% v/v) on the activity of purified mushroom polyphenol oxidase has been found to be as follows: (1)The oxidation of catechol or pyrogallol is greatly stimulated,an increase in the rate of oxygen uptake of over 100% having been invariably observed with catechol as the substrate,whereas the oxidation of other substituted o-dihydric phenols is inhibited in 50% ethanol. (2)When excess ascorbic acid is added to the reaction mixture to reduce o-quinone back to catechol as soon as the former is formed,the same stimulatory effect of ethanol on the rate of oxygen uptake has also been observed,indicating that ethanol stimulates only in the early stages of catechol oxidation.Treatment of the experimental results obtained in the presence of excess ascorbic acid and of different catechol concentrations according to Lineweaver and Burk shows that ethanol doubles the maximal velocity and raises the Michaelis constant 7-fold from 0.072 mM to 0.49 mM. Similar results have been obtained with pyrogallol as the substrate.This seems to suggest that the enzyme has a lower affinity for its substrates in 50% ethanol than in aqueous solution. (3)From simultaneous determinations of the amount of oxygen con- sumed and the amount of ascorbic acid oxidized the value of 1 is obtained for the ratio of oxygen consumed:ascorbic acid oxidized,both in the absence and in the presence of ethanol.This seems to indicate that the presence of ethanol does not affect the complete reduction of oxygen to water by the action of polyphenol oxidase. The effect of a number of other organic solvents on catechol oxidation has also been examined.Methanol,iso-propanol,tert-butanol and acetone are all stimulatory whereas dioxane,ethylene glycol and glycerol are in- hibitory.In 50% acetone,the rate of oxygen uptake is 3 times that of the control in aqueous solution.

    RNA synthesis during antigen recognition in rabbit peritoneal macrophage was investigated by incorporation of 3H-uridine in vitro.When the red cell membranes (RCM) from different sources were used as antigens,both autogenic and other RCMs from sheep,human,and allogenie rabbit stimulated the incorporation of 3H-uridine into RNA,but in different extents (allogenie rabbit > sheep,human > autogenic rabbit).When the normal transcription in macrophage was completely inhibited by rifampicin or actinomycin D,however,the effect of autogenic RCM was obviously different from those of the other RCMs: RNA synthesis in macrophage could not be stimulated by autogenic RCM anylonger,while the incorporation of 3H-uridine into RNA was still stimulated by other RCMs in the presence of rifampicin or actinomycin D.In order to characterize the RNA synthesized in antigen recognition,the synthesized 3H-RNA was fractionated by the 2.5% polyacrylamide -0.5% agarose gel electro-phoresis.It was shown that all membranes mentioned above stimulated the synthesis of several kinds of RNA,on the other hand,when normal transcription was completely inhibited by rifampicin,,foreign antigen only stimulated the synthesis of 4-5S RNA.The results of affinity chromatograph on poly(U)-Sepharose suggested that the 4-5S RNA carries poly (A).According to this results,it is suggested a work hypothesis about the process of antigen recognition in macrophage,which may be divided into two steps: primary and secondary recognition.The RNA synthesis related to antigen recognition in secondary step could be distinguished from those related to phagocytosis in primary step by rifampicin inhibition.The special 4-5 S RNA synthesized in secondary recognition may be catalyzed by a RNA polymerase which is DNA-independent but could not be inhibited by high concentration of rifampicin.The enzyme might present in a zymogen form and may only be activated by stimulation of foreign antigen.

Dosage of the antioxidant was increased by steps in three experimental groups at 0.04%—0.5%, 0.08%—0.6% and 0.24%—0.8%, respectively. All surviving experimental (63) and control (19) animals were sacrificed at the end of the 2nd experimental year and subjected to detailed anatomical and histological examination. Experimental mice showed 72.7% malignant tumors and 57.1% carcinomas, including cancers of various organs with those of the lungs, salivary glands, pancreas and kidneys predominating. There was dose-response relationship regarding incidence of carcinomas. Control mice had 21% sarcomas (mainly lymphomas) but no carcinoma. Results of the present experiment confirmed earlier findings of the carcinogenicity of antioxidant D in rats and suggested selective affinity of the carcinogen to the lungs as previously suspected.

    以含N-苯基-2-萘胺约85%,N-苯基-1-萘胺约4%且不含2-萘胺的国产防老剂D混于饲料中,饲喂幼年雄性C57/BL小鼠共一年半,饲料中防老剂D浓度在三个剂量组均逐步增加,分别为0.04%—0.5%、0.08%—0.6%及0.24%—0.8%。停止饲喂后半年杀死全部存活动物,作仔细的组织学检查。实验结果证明防老剂D对小鼠有致癌作用。63头实验组小鼠的恶性肿瘤总发生率为72.7%,癌瘤发病率为57.1%,其中主要包括肺癌、肝细胞癌和颌下腺癌,19头自然对照小鼠有21%肉瘤但无癌。实验和对照组癌发生率和肺、肝、颌下腺腺癌的各别发生率在统计学上均有显著差异。实验组中也较常见肝、脾、肾等血管内皮肉瘤的发生。肿瘤发生率和防老剂D的剂量之间存在着一定的相关性。
