The experiments were performed on 14 dc.gs and 11 rabbits anesthetized with sodium pentothal to determine the relationship between plasma hemoglobin(PHb) and disseminated intravascular coagulation (DIC) induced by endotoxin (ET) in dose of LD750 After ET injection platelet, plasma fibrinogen, prothrombin time, serum fibrin(ogen) degradation products and plasma protamine paracoagulation test showed the changes of DIC. PHb concentration elevated markedly at the same time. The changes of PHb was the second in order of abnormality percentage and the first according to the change ratio. Thus the determination of PHb concentration can serve as a supplementary index for DIC diagnosis. The fact that there was only an intermediate negative correlation between PHb and platelet count and fibrinogen concentration shows that microangiopathy hemolysis associated with DIC is one of the causes of the increase in PHb concentration and suggests that the impairment of unit membrane may also contribute to the changes of PHb concentration. As the mortality of the animals with DIC is in direct proportion to PHb concentration, it is possible that PHb concentration is regarded as an index to evaluate the severity and the prognosis of ET-induced DIC.

The human placenta was perfused to remove most of the blood, homogenized and extracted with Triton X-100. The thrombomodulin was purified from the extract with two chromatography of DEAE-sepharose and Thro-mbin-sepha-rose to obtained 0.25mg of thrombomodulin from 3.5 kg of placenta. The proatein was almost homogeneous as judged by electrophoresis on SDS-PAGE in teh presence of 2-mercaptoethanol with an apparent Mr=88000. Western blot anlysis without 2-mercaptoethanol gave an apparent Mr = 71 000. The resultin...

The proatein was almost homogeneous as judged by electrophoresis on SDS-PAGE in teh presence of 2-mercaptoethanol with an apparent Mr=88000. Western blot anlysis without 2-mercaptoethanol gave an apparent Mr = 71 000. The resultin isoelectric focusing showed thrombomodulin to be acid-protein, with p of 3.92~4.25. The purified thrombomodulin was inactivated by disulfide bond reduction, but was stable to pH extreme and protein denaturant. The thrombomodulin appears to have species-specificity in the inhibition of thrombin procoagulation as judged by different combination of bovine, human thrombi and fibrinogen. The thrombomodulin-thrombin complex greatly stimulated protein C activation by S_(2366) in vitro. The inhibition of protein C activationby antithrombin- Ⅲ was increasad with varying amount of antithrombin-Ⅲ. Thus, the protein purified is the thrombomodulin, a potent cofactor for protein C activation.
