The present study aimed to clarify the promising inhibiting effect of L-Glutamic acid on GA or GA+EC preserved porcine valves, and illustrate the processes of the leaflets surface covered by SD sucking rat's aortic endothelial cells. The porcine aortic valves were treated in following 4 groups: 1 ) 0. 5% GA treated valves, 2) none special biochemical treated valves, 3) valves treated by 0. 5% GA and then in 8% L-Glutamic acid solution, 4) 0. 5% GA for 72 hrs then 4% EC for 48 hrs before stored...

The present study aimed to clarify the promising inhibiting effect of L-Glutamic acid on GA or GA+EC preserved porcine valves, and illustrate the processes of the leaflets surface covered by SD sucking rat's aortic endothelial cells. The porcine aortic valves were treated in following 4 groups: 1 ) 0. 5% GA treated valves, 2) none special biochemical treated valves, 3) valves treated by 0. 5% GA and then in 8% L-Glutamic acid solution, 4) 0. 5% GA for 72 hrs then 4% EC for 48 hrs before stored in the solution of 8% L- Glutamic acid. Counting seeded-cells from valve leaflets; Identification of cellular type by factor Ⅷ rAG; and electronmicroscopic assessment after endothelial cells were seeded in 1, 3, 7, and 14 days. The result showed: All the leaflets of group 2, 3, and 4 could acquire the endothelial cells and fibroblasts covering during 7-14 days. The covering rate in group 2 exhibited more than 80% after 7 days but in group 1 no viable cells could be seen in 2-3 days after implanting. The study revealed that the toxicity of GA in the heterografts were inhibited markedly by L-Glutamic acid and their biocompatibility were improved that make sucking rat aortic endothelium could grow regularly on the surface of bioprosthetic valves.

我们将经过戊二醛(GA)和GA+环氧氯丙烷(EC)化学改性处理的猪主动脉瓣,再用L—谷胺酸处理以后进行SD乳鼠主动脉内皮细胞种植,并与单纯GA瓣及未经GA处理的瓣叶做对比观察。结果发现:单纯GA瓣种植细胞未能存活,而经L—谷胺酸处理瓣叶却获得种植细胞规律性生长、增殖,两周后完全覆盖瓣叶表面。电镜观察及第Ⅷ因子染色证实,瓣小叶表面生长的内皮细胞中夹杂着大量成纤维细胞生长。与种植前培育细胞纯化及种植以后成纤维细胞控制不够有关。

The secretion of the insulin of the microencapsulated purified neonatal porcine islets during culture was studied. However, the total energy loss (ELtot )of valves decreases with increasing HR,AP and VS,and the ELtot of tissue valves and Stars-Edwards valve increases with increasing CO,but Bjork -Shiley Monostrut and St Jude Medical valves have a higher ELtot at 2. 0 L/m and 8. 0 L/m of CO than at 4. 0 L/m and 6. 0 L/m of CO. In any condition ,for the tissue valve,Sorin-Pericarbon (SP)pericardial valve has a lower ELtot than Carpentier-Edwards (CE) porcine valve does;for the mechanical valve,the SE has a higher ELtot than Bjork-Shiley Monostrut (BS)and St Jude Medical (SJ)do except at 2. 0 L/m of CO and 100 bpm of HR.
