For the purpose of seting up a simple and effective method of cryopreservation of cord blood hematopoietic stem cells, a feasibility of cryopreservation and transplantation with cord blood hematopoietic stem cells stored in 4℃ refrigerator was investigated, indicating that it was appropriate that the time of cryopreserving the hematopoietic stem cells in cord blood at 4℃ should be 2~3 days and the recovery rate of cryopreserved remains almost unchanged. For the purpose of seting up a simple and effective method of cryopreservation of cord blood hematopoietic stem cells, a feasibility of cryopreservation and transplantation with cord blood hematopoietic stem cells stored in 4℃ refrigerator was investigated, indicating that it was appropriate that the time of cryopreserving the hematopoietic stem cells in cord blood at 4℃ should be 2~3 days and the recovery rate of cryopreserved remains almost unchanged. Using the 4℃ cryoprserved for 46 hours cord blood (nucleated cells: 1. 05 × 108/kg body weight), the successful sibling transplantation in a child with ALL was achieved and the bone marrow hematopoiesis was rebuilt. Reconstitution with donor cells was defined,which HLA typing and PCR-DNA finger-print detected donor's. With PCR detected, the specific TcR β band of T-lymphocytic leukemia was disappear. There are values of clinical application with cryopreservation of cord blood at 4℃.

为建立一种简便而有效的脐带血造血干细胞低温保存方法,我们探讨了4℃保存脐带血并用于临床移植的可行性。结果显示:脐带血4℃低温保存2~3天,造血干细胞回收率基本稳定;应用4℃保存46小时的脐带血(有核细胞数1.05×108/kg体重)移植治疗1例急性淋巴细胞白血病患儿,获得骨髓造血功能重建,有明确植活证据,并使白血病癌基因消失。说明脐带血的4℃保存具有一定临床价值。
