It hasbeen proved that retinoblastoma(RB)in some patients is associated withdeletion of one chromosome 13q ofsomatic cells and some data concern-ing the chromosomal behavior ofRB tumor cells reported in literatureare found to be similar to those con-cerning somatic cells. The aim ofthe present study was to analyzethe chromosome constitution of tu-mor cells and compare the charac-teristics of chromosome of the malig-nant cells with histological and clini-cal findings of the tumor. Results of chromosome study of so-lid tumor cells from 9 patients withRB are presented in this paper. Noneof the patients had a family historyof RB, and the examination of themetaphase of chromosome of peripher-al blood lymphocytes in 5 patientsrevealed nothing abnormal. Chromo-some preparations of the tumor cellswere obtained by using direct tech-nique and short-term culture (2 and24 h). Chromosome banding wasachieved in 4 cases only. The chromosomes in metaphase oftumor cells in all 9 cases were foundabnormal both in number and struc-ture. 30-206 metaphases of tumorcells were counted in each individualcase. The modal chromosome numberwas 45 in 3 cases, 46 in 2, 47, 66and 92 in 3 cases respectively, andhyper-tetraploidy in 1. Karyotypeanalysis revealed a considerable vari-ability. However, one to five markerchromosomes were found in each case.Six of these cases had aberrations ofD-group chromosome. Six of thesecases lost one E-group chromosome(chromosome 16) and had a metacen-tric chromosome in size similar to C-group chromosome [in some cases, itwas probably i(17q)].Four cases haddouble minutes. These results suggest-ed that the aberration of chromosomeD and E-group were probably relatedto the development and progression ofRB. The investigation of chromoso-mal, histological and clinical findingssuggested that some undifferentiatedtumors tended to have polyploid chro-mosome modes and the chromosomalpolyploidy was probably related toprotracted evolution of tumor cells inconsiderably long duration of malig-nancy too.

The Giemsa C-banding procedure was successfully modified to stain the nucleolus by prestaining the root tips with 1% aceto-carmine for two to three days before squashed. Four heterochromocenters appeared in the interphase cells correspondingly.With the band closely neighbouring the secondary constriction of chromosome Ⅵ (NOR-chromosome) as marker, it was found that both NORs in all the root tip cells were active and participated in nucleolus formation, and that uucleolus was exclucively associated with the banded NOR-chromosome (s) in mitotic as well as in interphaae cells.In cells with single nucleolus, the nucleolus was always associated with the pair of bands or heterochromocenters on the NOR-chromosomes but not with those on chromosome Ⅷ. It was clear that the single nucleolus was actually a fused body which was made up by the two NOR contributors.In cells with double nucleoli, it was observed that each nucleolus was always related with a single band or heterochromocenter on one of the NOR-chromosomes, but not with those on chromosome Ⅷ. It was apparent that one NOR-chromosome was responsible for one nucleolus formation in this type of cells. The one for one relationship provided us a direct explanation to Gates' dogma that the maxium number of nucleoli in a species was contant and equale to the number of secondary constrictions in the genome.It was also observed, in the cells with double nucleoli, that the two "nucleolus-band" structures were constructed in a polarized manner, with a band or heterochromocenter joining to the small end of a pearlike nucleolus. The two polarized structures were in a parallel distribution and oriented in the same direction inside the nucleus. This could be interpreted alterlatively by: 1) the chromosome arrangement within the interphase and prophasc still remained the telophase configuration in the mother cell; 2) the parallel distribution and the one-direction orientation could, with some unknown mechanism, have prevented the two polarized stuctures from fusing into a complex one.

The chromosome constitutions of several progeny from O.alta × O.sativa were analyzed with 22 mapped rice RFLP markers.The results showed that almost all triploid sterile plants which had 36 chromosomes were,as expected,composed of the chromosomes from two parents.But the two partial fertile regenerated plants from 02428(O.sativa)× WHS601 2(O.alta) which had only 24 and 25 chromosomes respectively,in most cases,had RFLP patterns similiar to their O.sativa parent. Therefore the chromosomes constituted their genomes were mainly from O.sativa or have high homology with the chromosomes of O.sativa. It seems evident that chromosome elimination had occurred during the wide hybridization as O.alta RFLP patterns could be found in them.Furthermore, that the introgression of O.alta chromosome fragment into that of O.sativa in an F 2 plant was indicative that some kind of recombination also occurred.Regarding the rare chromosome pairing in PMCs,the probability of the existence of a non conventional mechanism for the wild rice chromosomes (chromosome fragments )to introgressing into cultivated rice genomes in a relatively short time was discussed.
