It had been known that the lateraldiffusion coefficient(D) of the surfaceproteins of the mouse and sea urchineggs decreased markedly after ferti-lization by using fluorescencephotobleaching recovery (FRP) tech- ??nique,and accumalated into the clea-vage furrow during cytokinesis of mac-rophage and sea urchin egg.Usingthe FRP method we measured the mobilities of the soy bean agglutinin(SBA) and wheat germ agglutinin(WGA) receptors at the egg surface ofRana amurensis before and during thefirst cleavage....

It had been known that the lateraldiffusion coefficient(D) of the surfaceproteins of the mouse and sea urchineggs decreased markedly after ferti-lization by using fluorescencephotobleaching recovery (FRP) tech- ??nique,and accumalated into the clea-vage furrow during cytokinesis of mac-rophage and sea urchin egg.Usingthe FRP method we measured the mobilities of the soy bean agglutinin(SBA) and wheat germ agglutinin(WGA) receptors at the egg surface ofRana amurensis before and during thefirst cleavage. The animal pole of the denudedeggs scarcely had autofluorescence.Lectin receptors distributed more orless evenly (variation 20%) at thewhole animal pole when the denudedeggs were dyed with fluorescence labe-led SBA (50γ/ml) or WGA (10γ/ml).During cleavage,however,the lightintensity increased drastically alongboth sides of cleavage furrow wherethe folds emerged (Fig.1),and this wasconfirmed through point by point measurement which had the advantageto make the focusing more sharper. The FRP curves obtained beforeand during cleavage were of S-shape.Before cleavage some curves recoveredmore quickly,while others slowly(Fig.2a,2b).This might be relatedto the rhythmic contractions of eggs.According to Axelrod,the S-shapedcurve was casued mainly by flow.Ifit were caused wholly by diffusion,D would be 1×10~(-11) cm~2/sec as calculated from Fig.2b and the actual Dwould be much less than the abovevalue.This indicated that lectin receptors diffused very slowly. The polarity of the eggs being es-tablished after fertilization might simply be considered as a result of the flowof cytoplasm below the egg cortexrather than the diffusion of the cellsurface.During cleavage,the FRPcurves were more pronounced in S-shape when the light spots were placednear the folds presented at the sides offurrow (Fig.3).It was interesting to note that thelight intensity of the FRP curve mightbe stronger at the end of the experi-ment than that before bleaching (Fig.4) when the light spot was focusedonto the folds.This might be resulted from the emergence of increasing ??folding observed after the experiment.But,technically,it was very difficultto compare the increased light inten-sity with the increase of the numberand the height of foldings in the smallarea of light spot.The convex natureof FRP curve (Fig.5) then may beexplained by the result of loweringof the folding which could actuallyshow to increase the light intensity(Fig.1).As folds may emerge andvanish during the course of FRPmeasurement,the folding movementcould not be negelected in the analysisof the kinetics of FRP curve at leastin certain cases.

In SEM, sperm agglutination types vary with the kinds oflectin treated. The number, localization and possible functional significance of lectinreceptors on sperm membrane were briefly discussed.

取具有正常生育力男性的新鲜精液标本110例,彻底洗涤,以去除精子表面附着的精浆。调整精子浓度达15×10~6个/毫升,分别加入浓度为30微克/毫升的麦芽凝集素(WGA)、大豆凝集素(SBA)及刀豆球蛋白A(Con A),以721型分光光度计(500nm)测定从起始到20分钟期间的光密度变化值。结果表明,WGA、SBA及Con A均可引起人精子的明显凝集(与对照组相比,P<0.01)。我们认为,在恒定的实验条件下,可用此法衡量精子膜表面外源性凝集素受体的数目与功能状态。本文对110例标本由WGA引起的凝集反应值作了频数分布分析,为进一步研究在不同生理与病理条件下WGA受体的变化提供了基础数据。此外,还用扫描电镜观察了WGA、SBA与Con A引起的凝集类型,为确定外源性凝集素受体在精子膜表面的定位提供了依据。

Studies on the binding of FITC-TGL to ejaculated human spermatozoa have demonstrated the presence of lectin receptors all over the cell surface, but more in the arcosome region.The purified lectin were iodinated using the chloramine T method. The high specific activity (1.9 uCi//ug) 125I-TGL was used to bind human spermatozoa. The binding of TGL to its receptors is a saturation process with respect to the protein, and the scathard plot is nonlinear that suggested the conformation change of membrane components because of the presence of coopera-tivity. The maximum number of binding sites is about 4.1x102 per spermatozoon.
