OBJECTIVE:To apply the biosensor technology to screening of the best method to extract and separate anti-endotoxin effective materials in Radix Paeoniae rubra.METHODS:The affinities of samples extracted by three different methods binding to lipopolysaccride(LPS)were determined with biosensor technology.Extracted materials were incubated with0.25EU/ml endotoxin for determination of change of binding activity.RESULTS:Material extracted by water showed high binding capaˉbility with LPS,after incubation with endotoxin,still had highly effective anti-endotoxin component;1∶40diluted water extract could neutralize78.1%endotoxin.Determination results by biosensor technology and traditional limulus reagent showed no difˉference.CONCLUSION:Water extraction could obtain more anti-endotoxin effective materials.Compared with traditional methods,biosensor technology is a fast,highly effective,direct and accurate method.

AIM: To isolate and extract anti-endotox effective components in Radix Paeoniae Rubra. METHODS: By the biosensor technology, the anti-endotoxin effective components were isolated and extracted with general separate technology of Chinese traditional medicine. Through quantificational mensuration for LPS and inhibition of TNF-α release from endotoxin-stimulated cells in vitro, anti-endotoxin activity of Radix Paeoniae Rubra were elucidated. RESULTS: The effective components had high binding capability with LPS. After incubation with endotoxin and cells, the capability showed anti-endotoxin and inhibition of TNF-α release from endotoxin-stimulated cells in vitro. CONCLUSION: The anti-endotoxin effective components isolated and extracted using LPS as target by the biosensor technology are available in searching anti-endotoxin agents from Radix Paeoniae Rubra. It shows that Radix Paeoniae Rubra has high ability on neutralization with LPS.

Objective To study the anti-endotoxin effect of β-1,2,3,4,6-penta-O-galloyl-D-glucopyranose (PGG) in vitro. Methods The affinity of PGG with lipid A was determined with biosensor technology, and the endotoxin-neutralizing effect was assayed with LAL. Human peripheral blood mononuclear cells (hPBMC) were separated from healthy donors and cultured in vitro. The effect of different concentrations of PGG on the release of TNF-α and hIL-6 from LPS-stimulated hPBMC were measured by ELISA method. Results Lipid A was combined with different concentrations of PGG. The combination speed was shortened with the increase in PGG concentration. The KD value between PGG and Lipid A was 5.2×10~-7mol/L. The release of TNF-α and IL-6 of hPBMC under LPS stimulation( 958±234 ng/L vs 1 351±99 ng/L) was obviously inhibited by PGG in the concentration of higher than 20 mg/L compared with that without PGG treatment(1 788±171 ng/L vs 1 965±232 ng/L,P<0.05). Conclusion PGG show an anti-endotoxin effect in vitro, which may be associated with its ability to combine and neutralize endotoxin.

目的观察五没食子酰葡萄糖(PGG)对内毒素/脂多糖(LPS)的体外拮抗作用。方法应用生物传感技术测定PGG与脂质A(lipidA)的亲和力,动态浊度法测定PGG对LPS的中和作用。分离培养人外周血单核细胞(PBMC),观察不同浓度的PGG对LPS刺激下人PBMC释放肿瘤坏死因子(TNF)α、白细胞介素(IL)6的影响。结果不同浓度的PGG与脂质A发生结合反应,其反应的速率随浓度的增高而增快,二者的解离常数(Kd)值为5.2×10-7mol/L。PGG对LPS具有较强的中和作用。PGG在20mg/L以上的浓度时能够显著抑制LPS刺激下TNF-α和IL-6的释放,此时其TNF-α和IL-6水平分别从(1788±171)、(1966±232)ng/L降至(958±234)、(1351±99)ng/L(P<0.05),该作用具有一定的剂量效应关系。结论PGG可通过对LPS的结合及中和作用,在体外发挥拮抗LPS的作用。
