DNA content of 38 hepatocellular carcinomas (HCC) was quantitatively determined by means of image analysis technology .The results showed that DNA content in HCC was correlated well with the tumor size, capsule breaking, cancerous thrombus forming and clinical prognosis, that is, the higher DNA content was, the more malignance appeared, but not with the clinical staging and pathological grading of HCC. 66.7% of DNA diploid/near diploid HCC were less than or equal to 3cm in diameter, whereas 75% of DNA aneuploid HCC were more than 3cm in diameter (P<0.05), suggesting that HCC at 3cm in size may be an important period when the change of biological characteristics would occur. This study indicates that the quantitative analysis of DNA content can provide a valuable supplement to morphology in predicting the biological features and objective basis for assessing clinical prognosis of HCC.

本文报告应用图像分析技术,对38例肝癌的细胞核DNA含量进行了定量测定。结果显示;肝癌DNA含量与肝癌瘤体大小、包膜突破、癌栓形成以及临床预后有明显相关性,但与临床分期及病理分级无关;66.7%的DNA二倍体/近二倍体(D/ND)肝癌瘤体直径≤3cm,而75%的DNA异倍体(AN)肝癌瘤体直径>3cm,提示肝癌直径在3cm时可能是其生物学特性转变的重要时期。本文认为,对肝癌DNA含量的定量分析,可为从形态学上估价肝癌的生物学特性提供有价值的补充,并为判断临床预后提供客观依据。

The effect of cumene hydroperoxide on cultured endothelial cells (ECs) from human umbilical vein was investigated both morphologically and functionally. The effect of cumene hydroperoxide on cultured endothelial cells (ECs) from human umbilical vein was investigated both morphologically and functionally. The results showed that cumene hydroperoxide stimulated and faciliated the lipid peroxidation in ECs. The concentration of lipidperoxide in ECs increased with the increase of cumene hydroperoxide. At the same time, the ultrastructural changes were contraction, plasma membrane blebbing, dilatation of endoplasmic reticulum and mitochondria, mainly leing the changes of biological membrane. The concentration of 6-keto-PGF_1α in the medium increased slightly in lower concentration group of cumene hydroperoxide (0.1mM), whereas it decreased remarkably in the higher concentration group. It illustrated that the biosynthesis of prostacyclin was inhibited.

本实验用氢过氧化枯烯作用于培养的人脐带静脉内皮细胞,探讨脂质过氧化对血管内皮细胞的作用。结果表明,氢过氧化枯烯激发和促进了内皮细胞的脂质过氧化过程,使内皮细胞的过氧化脂质含量升高。在形态结构上内皮细胞出现收缩和生物膜系统的损伤。用放射免疫方法测定,内皮细胞培养液中的6-keto-PGF_1α含量减少,表明PGI_2的产生受到抑制。上述变化可认为:内皮细胞对脂质过氧化的作用是敏感的,提示脂质过氧化通过损伤内皮细胞,可能在动脉粥样硬化发病机理中起重要作用。

本文报道应用图像分析技术,对38例肝癌细胞核DNA含量进行定量测定。结果显示:6例为DNA干系二倍体,32例为DNA干系异倍体。肝癌DNA干系水平与肝癌瘤体大小、包膜突破与癌栓形成发生率以及临床预后等生物学特性有明显相关性,而与临床分期、血清AFP含量、病理分级、组织学类型无关。二倍体肝癌中,66.7%的瘤体直径≤3cm,而异倍体肝癌中,75%的瘤体直径>3cm(P<0.05),提示肝癌于3cm大小时可能是其生物学特性发生转变的重要时期。本文结果表明,对肝癌DNA含量的定量分析,可为临床估价肝癌的生物学特性和判断临床预后提供客观而定量的依据。
