Aqueous extractions of seventy-eight traditional Chinese herbs were tested using affinity biosensor technology. Then aqueous extractions of the herbs with high affinity for CpG ODN were incubated with CpG ODN(33 μg·ml -1)at 37 ℃ for 30 min, subsequently the mixtures were tested using affinity biosensor technology. RESULTS[WTBZ]: Among the seventy-eight Chinese traditional herbs, fourteen herbs such as radix paeoniae rubra, rhubarb, and terminaliae immaturus possessed the higher binding activity to CpG ODN, and nine herbs such as Java Brucea fruit, radix paeoniae rubra, and radix sanguisorbae possessed larger amount of effective binding-CpG ODN components. CONCLUSION: Nines kinds of Chinese traditional medicines possess the highly content components with highly specifically affinities to CpG ODN and it provides experimental base for further separation anti-inflammatory component from Chinese traditional medicine. [WTHZ]

目的:筛选具有拮抗CpGODN作用的抗炎中药。方法:将CpGODN包被于生物传感器的生物素化样品池上,测定78种中药水煎液与CpGODN的亲和力;选择具有较高亲和力的中药水煎液,与CpGODN(33μg·ml-1)37℃孵育30min后,再次测定与CpGODN的亲和力。结果:78种中药中,地榆、大黄、藏青果等14种中药含有结合CpGODN的活性成分;鸦胆子、赤芍、地榆等9种中药活性成分含量较高。结论:筛选得到9种活性含量较高的中药,为下一步分离提纯有效成分单体提供了实验依据。
