This paper has reported a method for the determination of glucose in the supernatants of microencapsulated animal cell perfusion culture using glucose biosensor. The glucose biosensor shows a good linearity of glucose concentration between 10 to 500 mg/dl with γ=0.999,which made it possile to use 20μl for each assay.Its response time is 40 seconds and it possesses high operational stability.The glucose oxidase membrances are stable at 4℃ for 40 days.

本文提出了用葡萄糖传感器监测在灌注培养微囊化动物细胞时,培液中葡萄糖含量的变化。实验过程中,葡萄糖传感器的线性检测范围为10~500mg/100ml。将葡萄糖氧化酶膜贮存于4℃条件下的0.05mol/LPBS(pH7.0)中,它的活力在40天之内保持不变。本方法操作简便,灵敏度高,耗时少,准确可靠。

A new method was developed to covalently immobilize enzyme onto the cellulose acetate (CA) membrane. 采用共价法将酶固定在醋酸纤维素膜上,方法简便易行,制造的酶膜稳定,比活力高。同时采用该方法制备了葡萄糖氧化酶酶膜,与氧电极组装成测定葡萄糖的生物传感器,线性范围为50~800mg/dl,仪器工作的最适pH为6.0,最适温度为40℃。将该膜与过氧化氢电极组装得到的传感器具有以下特性:线性范围为10~200mg/dl,最适pH为6.0,测定结果与酶试制盒有良好相关性。
