Characteristic of the biosensor has been shown by cyclic voltammetry and stationary potential measurements. The biosensor responds rapidly to glucose,the steady-state current response achieved in less than 50 s. The response to glucose under N2 saturation shows apparent Michaeclls-Menten constant Kμapp 11. 6mmol/L. The biosensor can be operated in a potential range where the oxidation of common interferingspecies, such as ascorbic acid and uric acid, does not occur. The lifetime of the biosensor is 30 d and plasmaglucose assays were correlated with spectrophotometry(Y=-7. 1 + 1.09 X,r = 0. 990, n = 8).

A new enzyme glucose sensor concept featuring glucose oxidase immobilized on fine carbon powder in a fluid state and a cross-linked glucose oxidase enzyme layer has been developed. This glucose biosensor has been tested in vitro at 37℃ and has a lifetime of three months after which it can be recharged with fresh enzyme.In the present study, further in vitro characterization of this sensor has been carried out. Most types of glucose sensors are susceptible to interence by body chemicals. The sensor response is thus nonreproducible and calibration is difficult. We have conducted interference tests on this biosensor using cellulose acetate, polytetrafluoroethylene (PTFE) and polycarbonate membranes.Some of these membranes were coated with positive or negative charge-carrying hydrogel layers. All sensors showed a stable and linear response to glucose concentrations up to 300mg/dL, in the presence of glucose alone in the phosphate buffer medium, as well as in the presence of seven interferences (used in partial factorial experimental design using two-, four-, and six-chemical combinations ) .The effects of ascorbic acid, bilirubin, creatinine, L-cystine, glycine, uric acid and urea on the amperometric signal of the sensor were studied, by use of a 2 ̄7 half-replicate factorial experimental design. A novel analytical method consisting of comparisons of slopes of the sensor calibration curves with and without interences has been developed. A statistical analysis of variance was performed on the slope changes thus obtained, and significance levels of the main effects (control), two-factor, four-factor and six-factor interactions were determined. All the membranes coated with charged hydrogels were partially successful in limiting interferences. The polycarbonate membrane coated with the negatively charged hydrogel was the most effective in decreasing the interferences, especially of ascorbic and uric acid.
