In current clamp mode, action po-tentials were recorded at current stimulation of 0.5Hz, 3 ms. Tet 30 μmol·L-1 shortened APD5072.2%, APD90 44. 0%, but had no significant effectson rest potential and action potential amplitude.From a holding potential of -40 mV, depolanzingpulses to a test potential (-30 mV to 60 mV) for 150ms at 0.5 Hz frequency evoked L calcium currents.The inward currents could be completely blocked by20 μmol· L-1 nifedipine and enlarged by 5 μmol·L-1 Bay K 8644. Tet, 0.3 to 300 μmol· L-1, inhib-ited calcium currents without changing the I-V rela-tionship. The inhibition of inward currents was con-centration dependent. The concentration forhalf-maximum inhibition was about 13.3 μmol· L-1.The inhibition of Ca2+ inflow was the major mecha-nism of antihypertensive and antiarrhythmic effectsof Tet.
