It was reported that Gypenosides is supposed to be effective in the protection of my-ocardial ischemia-reperfusion injury in rats and hippocampal delayed death (DND) following transient brain ischemia in gerbils.The purpose of this study is to observe the effect of Gypenosides on experimental cerebral ischmia-reperfusion injury. In this paper, Wistar rats were used. The cerebral ischemia-reperfusion injury model was produced by clipping basilar artery and bilateral common carotid arteries for 60 minutes and then reopenning three vessels to observe 60 minutes. Gypenosides were injected intraperitoneally in 100mg/kg body weight in 30 minutes before clipping three vessels. Observation was performed in proctive effect of Gypenosides on cerebral ischemia-reperfusion injury in rats. The results showed that cerebral coeffients(x ± s ) in control group,cerebral ischemia-reperfusion group and Gypenosides group were 4. 51±0. 33,5. 23±0. 18 and 4. 37±0. 45,respectively. Compared with ischemia-reperfusion group,cerebral coeffient in Gypenosides group was significantly lowered ( P < 0. 01). Cerebral water contents in control group,cerebral ischemia-reperfusion group and Gypenosides group were 78. 31 ±0. 00,80. 69± 0. 06 and 78. 60±0. 00(%) respectively. Compared with ischemia-reperfusion group,cerebral water content in Gypenosides group was significantly lowered( P <0. 05).Malondialdehyde (MDA) contents in rat brain in control group,cerebral ischemia-reperfusion group and Gypenosides group were 5. 15±0. 58,6. 39±0. 66 and 4. 60± 1. 80(nmol/mg wet weight)respectively. Compared with ischemia-reperfusion group,malondialdehyde (MDA) content in Gypenosides group was significantly lowered( P <0. 05). These results indicated that Gypenosides is effective in the protection of cerebral ischemia-reperfusion injury in rats.

本文观察绞股蓝总皂甙(100mg/kg)对大鼠全脑缺血再灌流损伤的作用。结果发现:脑系数对照组为4.51±0.33,再灌组为5.23±0.18,绞股蓝组为4.37±0.45;再灌组高于对照组,绞股蓝组低于再灌组,两两比较均有高度显著性差异(P<0.01 )。脑含水量对照组为78.31±0.00,再灌组为80.69±0.06,绞股蓝组为78.60±0.00(%);再灌组高于对照组,绞股蓝组低于再灌组,两两比较均有显著性差异(P<0.05)。脑组织丙二醛含量对照组为5.15±0.58,再灌组为6.39±0.66,绞股蓝组为4.60±1.80(nmol/mg湿重);绞股蓝组低于再灌组,差别有显著性(P<0.O5)。上述结果表明,绞股蓝总皂甙对大鼠脑缺血再灌流损伤有明显的保护作用。

The transient brain ischemia was produced in rat with method of four vessels occclusion. Changes of lipid peroxidation (LPO). superoxide dismutase (SOD) and acetylcholine (Ach) in rat cortex were sbserved after 20 min. of brain ischerrda and during three phases of reperfusion. The results showed that in period of reperfusion, the LPO in cortex increased;and the Ach rose Up as well. However. there was not obvious change in the content of SOD. This results may indicate that the reperfusion injury following ischemia is closely related to the increase free radical generation which may not be due to the damage of the radical clearing system of brain. In addition, the reperfusion injury also involved to the cortical cholinergic system.

本文采用四血管结扎法观察了大鼠全脑性缺血后血流再通三个时相脑皮质过氧化脂质(LPO)、超氧化物岐化酶(SOD)及乙酰胆碱(Ach)含量的变化。结果显示:血流再灌注期间脑皮质LPO明显增加,而SOD无明显变化,Ach的含量增加。说明脑缺血后血流再通所发生的再灌注损伤与氧自由基产生增加有关,而且可不伴有皮质对自由基清除系统功能的受损。同时说明再灌注损伤过程也牵涉皮质的胆碱能系统。

After gerbils were killed. the hippocampal CA1 neurons were numbered. SZ-1 was given ip at 10 mg/kg 30 min before ischemia.Results:The survival numbers in the ischemia group were decreased very significantly (ftom 153.5±1. 6 to 37. 4±11. 7, P<0.01) and 10 mg/kg SZ-1 increased the survival numbers of CA1 neurons ftom 37.4±11. 7 to 134. 8±7. 4 (p<0.01). Conclusion : SZ-1 has good protective effects on gerbil brain ischemia.

目的:研究新合成的血小板激活因子受体拮抗剂反式-2,6-双(3,4二甲氧苯基)-四氢吡喃(SZ-1)对沙土鼠脑缺血的保护作用。方法:结扎沙土鼠两侧颈总动脉造成短暂性脑缺血,缺血时间为10min,再灌注时间为5d。处死后计数沙土鼠海马CA1区750μm长度内神经无数。在缺血前30min腹腔给SZ-1(10mg/kg),比较假手术组、缺血组及给药组存活的神经元数。结果:缺血组神经元的存活数较假手术组非常显著的减少(从153.5±1.6下降到37.4±11.7,P<0.01),10mg/kgSZ-1可使神经元存活数显著增加(从37.4±11.7上升到134.8±7.4,P<0.01)。结论:SZ-1对沙土鼠脑缺血引起的神经损伤具有良好的神经保护作用。
