Using the technique of combined Feulgen microspectrophotometry and ~3H-thymidine autoradiography,it has been found that in early larval life of Culex pipiens pallens,the majority of sali-vary gland cells which do not incorpo-rate ~3H-thymidine have DNA contents which fit a 2~n geometric series,when Feulgen-DNA estimates for the me-taphase chromosomes of spermatogo-nium are used as the base for computing such series.Table I shows that the DNA C-values of Culex pipiens pallens is 0.585 picogram...

Using the technique of combined Feulgen microspectrophotometry and ~3H-thymidine autoradiography,it has been found that in early larval life of Culex pipiens pallens,the majority of sali-vary gland cells which do not incorpo-rate ~3H-thymidine have DNA contents which fit a 2~n geometric series,when Feulgen-DNA estimates for the me-taphase chromosomes of spermatogo-nium are used as the base for computing such series.Table I shows that the DNA C-values of Culex pipiens pallens is 0.585 picogram DNA and table II shows the percentage distribution of the different kinds of salivary gland nuclei at different stages of larval develop-ment.Based on the relative content of DNA and the incorporation of ~3H-thymidine,three kinds of cells can be classified: (1) ~3H-thymidine labelled cells in S phase with DNA contents fall mainly between 2~(n+1)C and 2~(n+1)C.(2) Unlabelled cells in G phase with DNA contents fall within the multiples of 2~nC and (3) Unlabelled cells between the polidy stages or U-cells with DNAntents fall also between 2~nC and 2~(n+1)C.In the late fourth larval instar,more than the half of the cells of salivary gland are unlabelled U cells,whose DNA contents deviate from 2~n series.Figure 3 also shows the histograms ofthe relative DNA contents,spread on alogarithmic scale and ~3H-TdR label-ling (dark areas) of salivary cell nucleiat different larval instars.Meanwhiles,Feulgen-stained squashed salivary glandchromosomes prepared from thesestages also showed an accumulation ofFeulgen-positive material in some chro-mosomal segments.These facts takentogether indicated that differentialDNA replication occurred during thepolytenization of Culex pipiens pallenssalivary gland chromosomes. Besides,the autoradiography alsoshowed that the labelled cells can bedivided into two patterns:continuouslabel and discontinuous label.Cytopho-tometric determination of DNA contents of these two pattens showed thatthe continuous label represented earlyand middle S phase and the discontin-uous label represented late S phasehich synthesize only a minor amount of DNA.The distribution of these two types of cells in the third instar represented in figure 2,in which theFeulgen-DNA relative absorption (R.A.) of continuous (●) discontinuous(○) labelled nuclei and unlabellednuclei (×) are plotted against thedistribution of nuclei.

(1)应用DNA—孚尔根细胞光度术与。~3H-TdR放射自显术相结合的方法分析每一细胞,发现在库蚊幼虫早期发育中,唾液腺细胞核DNA含量基本上呈2~n倍数增加。但在各龄期中均有一定比例的U细胞,其DNA含量虽偏离2~n倍数,但并非正在合成DNA;四龄后期此种细胞可达60%以上,并相应地在染色体上不同区域观察到DNA的不同积累,表明在库蚊幼虫唾液腺细胞核内DNA存在不均等复制。(2)用细胞光度法测定了库蚊精原细胞,脑神经节细胞分裂中期染色体和精子DNA含量,算得每单倍体DNA含量0.568微微克。以此为确定倍数值的基数,显示一龄幼虫唾液腺细胞核DNA含量主要为16C,二龄居于16C—32C之间,三龄64C,四龄居于128C—256C之间,最高倍数值860C。(3)放射自显术显示,连续复制居于S期的前期和中期,合成染色体大部分DNA;不连续复制居于S期的后期,合成的DNA所占比例较低。

The action of EMD on DNA content in lung cancer A-549 cell strain was detected by means of Leits MPV3 microscope photometer. It was found that DNA content in A-549 cells treated with EMD was decreased markedly. The peak appeared in the low channel and moved to the left. It is suggested that EMD has an evident restriction on the division and growth of A-549 cells.

用Leitz MPV_3型显微分光光度计研究了EMD对肺癌A-549细胞株DNA含量的影响,发现EMD作用后肺癌细胞的DNA含量明显减少。测定单个细胞中DNA的含量可以获得细胞在各个细胞周期分布的资料。EMD浓度为10μg/ml时在第3和第4天引起G_1和S期细胞增多,G_+M期和高倍体细胞减少,高峰在低通道处出现并左移,表明EMD对肺癌A-549细胞的分裂增殖有抑制作用。

This report presents the study of determination of DNA in different Namalva IFN preparation by a microassay method using ethidium bromide. The results show that the purified IFN,by antibody affinity chromatography, contains no DNA, which indicates this purified IFN can be used clinically as a safe drug.

本文报道了用溴乙锭DNA微量测定法对不同Namalva IFN制品中DNA测定的研究。结果表明,经抗体亲和层析纯化的IFN不含有DNA,从而证实该纯化IFN制剂能够作为安全制品用于临床。
