The present investigation deals with the plantlet formation induced from the stem segments of Buddleia asiatica Lour,in vitro and the effect of plant hormones on the callus growth and organogenesis in tissue culture. The present investigation deals with the plantlet formation induced from the stem segments of Buddleia asiatica Lour,in vitro and the effect of plant hormones on the callus growth and organogenesis in tissue culture. It was found that plantlets were obtained on MS basic medium sup-plemented with BA (2mg/l) plus NAA (0.2 mg/l) for callus induction,BA (1.0mg/l) for bud differentitiation,and 1/2 MS basic medium with the addition of IBA,IAA,NAA or 2,4-D at the same concentration (0.5 mg/l) for root formation.Among the auxin tested,NAA was more superior to any others in rooting and 2,4-D showed no activity on root formation. When the calli induced from stem segments,they were transfered to the redifferentiation media,MS medium with the addition of varying concen-tration of BA and NAA,the callus growth and organogenesis were regula-ted by quantitative shifts in the ratio between those two hormones.The re-sults obtained showed that: 1.Exo-genous application of a relatively low concentration of BA and NAA was indispensable for callus growth on the culture.2.BA supplemented alone greatly stimulated bud formation in concentrations ranging from 0.25-4.0 mg/l,but suppressed completely root induction; whereas NAA supplemented alone markedly accelerated root for-mation in concentrations ranging from 0.5-6.0 mg/l,but suppressed bud in-duction with increasing concentra-tions.3.In the presence of varying concentrations of BA and NAA in combination,a cytokinin-auxin ratio greater than 0.5 was found to be fa-vourable for bud formation but sup-pressible for root induction; whereas a cytokinin-zuxin ratio lower than 0.5 was found to be favourable root for-mation but inhibitory to the shoot promoting effect of cytokinin.The counter actions between auxin and cytokinin in these respects have been briefly discussed.

在驳骨丹离体茎切段培养中,用MS基本培养基,分别附加2毫克/升BA和0.2毫克/升NAA诱导愈伤组织,附加1毫克/升BA诱导芽分化;以2/1MS基本培养基分别附加0.5毫克/升NAA、IBA、IAA和2,4-D进行诱导发根试验,除2,4-D无效外,均能生根形成再生植株。根据上述试验结果,设计不同浓度的BA和NAA组合,进行愈伤组织继代培养,讨论这两种激素对愈伤组织生长和器官再生的调节作用。主要结果为:1.低浓度的BA和NAA组合比较有利促进愈伤组织的生长,且以中等浓度的BA和低浓度NAA的效果最佳;2.单独附加BA,其浓度在0.25—4.0毫克/升范围内,都能促进芽分化,而完全抑制了根的发生。当单加NAA,在0.5—6.0毫克/升的浓度范围内,能促进根的形成,而随其浓度的升高抑制芽的分化;3.细胞分裂素与生长素的比值>0.5时,有利芽分化而抑制根的发生,当比值<0.6时,有利根形成而抑制或减弱细胞分裂素促进芽分化的作用。在这里讨论了生长素和细胞分裂素之间对报、芽形成的拮抗作用。

本文报道了山茶科金花茶系七种会花茶的组织培养快速繁殖育苗技术,研究了在改良的ER培养基中~[1],七种金花茶在胚状体形成,假珠芽成苗~([6,7]),愈伤组织分化芽及用成年树茎尖进行腋生株快速繁殖等再生方式中的不同效应,试验证明:金花茶系植物同其他山茶科植物一样,其体细胞能通过多种再生方式产生大量的完整植株。本试验所用的七种金花茶无论在所用分化配方,再生方式及移栽方法上都与我们做过试验的茶树,油茶,山茶花等多种山茶科植物极其相似,因此本试验进一步证明可分化配方与种属密切有关,而且在分化情况及再生方式方面都具有明显的共性,我们根据此一结果正把此方法用于金花茶杂交幼胚的早期培养上,现已得到大量的杂交小金花茶苗。

为有效筛选烟草抗赤星病细胞突变体,本研究建立了一种双层培养基筛选系统。即在下层培养基中接种烟草赤星病(Alternaria alternata),在上层培养基中接种烤烟品种 NC89和净叶黄的花药(NC89高感赤星病,净叶黄高抗赤星病)。在26±1℃培养40天后发现,NC89的花药出苗率和花药平均出苗数与对照(不接种赤星病的培养基)相比,分别由30%和53颗降到2%和12颗,而净叶黄的花药出苗率和平均出苗数基本没有变化。对筛选出来的 NC89花培突变体进行了田间赤星病抗性鉴定,其中80%对赤星病抗性较 NC89有所提高,甚至部分接近高抗。这说明该双层培养基系统可以有效地对抗烟草赤星病的细胞突变体进行筛选。本系统尤其适应于(1)需长期培养;(2)毒素不易制备或易失活的病原菌的抗性筛选。
