On the other hand, methanol pretreatment had no effect on hepatic MFO activity,especially cytochrome P-450 and cytochrome b5, but it could significantly reduce the SH-radical amount of liver in rats.Therefore it might be one of the possible me chanisms of methanol potentiation.

本文选用大鼠研究了甲醇对CCl_4肝毒性的加强作用,其程度随剂量的不同而有很大差异,最大加强作用的时间为36—48小时。甲醇预处理能引起体内、体外脂质过氧化作用,但对CCl_4引起的脂质过氧化无明显加强作用。另一方面,甲醇预处理对鼠肝MFO活性,特别是对Cytp_(450)及Cytb_5无明显影响,但能明显降低大鼠肝脏巯基含量,因此,可能是甲醇加强作用的机理之一。

The effects of several traditional Chinese medicinal herbs on TNT-induced hepatotoxicity were investigated in acute., subchronic TNT-treated rats. It was found that Buplerum chinense DC, Schizandra chinesis ( Turcz ) Baill, Glycyrrhiza uralensis Fisch and Rehmannia glutinosa ( Gae rth ) libosch each has an inhibitory effect on TNT-induced enhancement of hepatic TBA reaction products ia acute TNT-treated rats and the decoction made of these four herbs exerted an inhibitory effect on TNT-induced decrease of serum ceruloplasmin amount in subchronic TNT treated rats. But the herbs selected showed no effect on TNT-induced enhancement of hepatic phospharylase a activity in acute TNT-treated rats and TNT-induced changes of hepatic TBA reaction products, hepatic GSH amount etc. in subchronic TNT-treated rats.

研究结果发现,柴胡、生地、五味子、甘草对急性染毒大鼠肝脏TBA反应物的升高和这4种中药制成的混合剂对亚急性染毒TNT大鼠血清铜蓝蛋白含量的降低有一定抑制作用。但所选六种中药对急性染毒时肝脏磷酸化酶a活性的升高和亚急性染毒所致肝TBA反应产物、GSH含量等的改变无明显拮抗作用。

采用Sephadex G-75凝胶层析技术对经口染毒亚砷酸钠的大鼠肝内砷结合物进行了分离和分子量测定,并对砷在肝脏亚细胞组份中的分布作了初步探讨。结果表明:95.7%的砷分布于细胞质中,4.3%的砷在细胞核中。细胞质中的砷与分子量45,000,24,000道尔顿的蛋白质结合及分子量6000的小分子物质结合,其中蛋白质结合砷占68%,小分子砷结合物占32%。两种砷结合蛋白与小分子砷结合物在砷中毒肝损害中的毒理学意义值得进一步研究。
