We investigated the expression of TGF-α in mouse Ehrlich ascites carcinoma cells from 5 th and 12th day after inoculation.TGF-α was detected by radioimmunoassay and the TGF-α-mRNA was identified with spot hybridization method. The results show that there was expression of both TGF-α immunoactive molecule and TGF-α-mRNA in mouse Ehrlich ascites carcinoma cells.The intensity of expression in mice after inoculation 12 day was more than that after 5 day.

P 15 INK4B gene was frequently methylated in patients with high risk MDS and early stage of MM. Conclusion Hypermethylation of P 15 INK4B gene is one of the main causes of its inactivation. Hypermethylation of CpG island was closely related to the development of malignant hematopoietic diseases.

Objective To explore gene expression of vascular endothelial growth factor(VEGF) and its receptors in human hematopoietic cells.Methods Total RNA was isolated from mononuclear cells of peripheral blood(PB) and bone marrow(BM) as well as from hematopoietic tumor cell lines.Expression of VEGF and its receptors,Flt 1 and KDR,were analyzed using reverse transcriptase polymerase chain reaction(RT PCR).The expression level was normalized by β 2 microglobulin gene.Results VEGF transcripts were detected in PB from 18 of 22 donors,BM from 19 of 22 non malignant cases and all of the 10 tumor lines,respectively.Flt 1 gene expression was confirmed in 4 of 12 donors in PB,in 9 of 22 cases in BM and in 6 of 10 tumor lines.Fourteen of 22 BM samples in addition to Raji cells were found to express KDR at a low to moderate level,but not in PB.Moreover,platelet also expressed VEGF and Flt 1 receptor.Conclusion These findings suggest that VEGF,an effector of angiogenesis,may play an important role in growth of hematopoietic cells,especially in hematopoietic malignancies.

目的 探究血管内皮生长因子 (VEGF)及其受体Flt 1和KDR基因在造血细胞中的表达。方法 提取正常外周血、骨髓单个核细胞及血液肿瘤细胞系的总RNA ,以 β2 微球蛋白基因为内标 ,用半定量逆转录聚合酶链反应 (RT PCR)分析VEGF ,Flt 1和KDR基因的表达。结果 在 2 2份正常外周血和 2 2份非恶性骨髓标本中 ,分别有 18份和 19份表达VEGF基因 ,而 10种人类血液肿瘤细胞系均有VEGF基因的高度表达。在 12份外周血中 ,有 4份检测到Flt 1基因表达 ,但均未见有KDR基因表达 ;在 2 2份骨髓中 ,分别有 9份和 14份表达Flt 1基因或KDR基因。 10种血液肿瘤细胞系有 6种表达Flt 1基因 ,而KDR基因表达仅见于Raji细胞。血小板也有VEGF和Flt 1基因的低度表达。结论 VEGF作为血管生成的介导物 ,可能具有调节正常造血细胞和血液肿瘤细胞生长的作用
