The positive rate of IgM anti-HD was 4.9%.This method is significant in early diagnosis and epid-miological study of hepatitis D.

ObjectiveTo explore the expression and relationship of hepatitis D virus antigen (HDAg)/hepatitis delta virus ribonucleic acid(HDVRNA) and hepatitis B virus deoxyribonucleic acid(HBVDNA) in liver tissue of the patients with hepatitis D(HD)Methods HDAg,HDVRNA and HBVDNA in paraffinembedded liver tissue from 79 liver biopsy or autopsy samples from the patients with HD were studied with immunohistochemical techniques and hybridization in situ,meanwhile,compared with the expression of HBVDNA in liver tissues of the patients with hepatitis B(HB)ResultsIt showed that the detection rate of HBVDNA in liver specimens of HD(27%,21/79) was lower than that of hepatitis B(44%,23/52)(P<005).Strong expression of HDAg and HDVRNA were shown in the bollooning degeneration hepatocytes and hepatocyts near piecemeal necrosis region.HDAg expression was significently negative association with HBVDNA expression(P<005).ConclusionHDV infection can inhibit replication of HBVDNA,espcially at strong expressiong of HDAg or HDVRNA. Besides interactions between HBV and HDV,it is possible that direct cytotoxicity of HDV may play an important pathogenetic role in pathogenetic mechanisms of HDV. [

目的探讨丁型肝炎病人肝组织中 HDAg、HDVRNA与 HBVDNA表达及关系。方法应用免疫组化和原位杂交技术检测了 79例丁型肝炎病人肝组织 HDAg、HDVRAN、HBVDNA表达 ,以 5 2例乙型肝炎病人肝组织 HBVDNA表达作对照。结果丁型肝炎 HBVDNA检出率 ( 2 7% )低于乙型肝炎 ( 44 % ) ( P<0 .0 5 )。在坏死灶边缘肝细胞和气球样变肝细胞浆内有大量的 HDVRNA蓄积或 HDAg呈浆型强表达 ,HDAg与 HDVRNA表达及分布呈一致性 ( P>0 .0 5 )。HBVDNA与 HDAg表达强度呈负性相关 ( P<0 .0 5 )。结论 HDV感染会抑制 HBV复制 ,尤其是 HDAg或 HDVRNA强表达时 ;在 HDV致病机制中可能既有 HDV的直接细胞毒性作用 ,也有 HBV和 HDV的协同作用

Objective:To study the effects of hammerhead ribozymes delivered by hepatitis delta virus(HDV) targeted on hepatitis B virus(HBV),in cell culture and an animal model,on the HBV gene expression and replication.[WTHZ]Methods:Based on the construction of a co-expression plasmid of replication-competent HDV-ribozyme and HBV, the effects were severally tested by transfection of Huh-7 cells and hydrodynamic injection of mouse model in vivo. Results:It was suggested that inhibition induced by recombinant HDV-ribozyme on HBsAg expression is relative to both the recombinant site of HDV and the ribozyme target.Hydrodynamic injection of the viral over-length genome leaded the expression of recombinant HDV and HBV.According to the cell culture system,the recombinant HDV-ribozyme exhibited effectively inhibition of HBV gene expression and replication in the livers and sera of the treated mice.Conclusion:This inhibition experiment in vivo represents a basis for further construction of therapeutic recombinant HDV,and then the development of targeting antiviral gene therapy.

目的:研究用丁型肝炎病毒(HDV)作为载体携带乙型肝炎病毒(HBV)特异性的锤头状核酶所构建的重组体,在细胞体系及转染动物模型中对HBV基因表达和复制的影响。方法:将HDV核酶重组体和HBV的共表达质粒转染Huh7细胞以分析HDV核酶重组体对HBV基因表达的影响;用小鼠尾静脉快速注射法将共表达质粒转染到小鼠体内,检测重组体在动物体内对HBV基因表达和复制的抑制作用。结果:转染细胞中,重组体对HBsAg的抑制与HDV重组位点和核酶靶位都有关;水压法注射的质粒在小鼠肝内得到表达,与对照相比重组HDV核酶可有效抑制在肝和血清中HBV的基因表达以及复制,与细胞中的结果一致。结论:此项体内实验为进一步构建治疗性重组HDV病毒,发现靶向性抗病毒基因治疗手段奠定基础。
