We believe that the platelet hyperactivity is an important factor in the pathogenesis of actrte cerebral ischemic stroke. In patients with long-term hypertension, the increase of platelet aggregation may be considered as a possible risk factor in the development of cerebral thrombosis and transient ischemic attack.

We have used 14 mongrel dogs to establish the experimental cerebral infarction of MCA since 1984. The proximal segment of the right MCA and distal segment of the ICA were clipped. We have observed and recorded the clinical findings of each dog every day, according to Crowell and Olsson's five scales. The postoperativc surviving animals were sacrificed 24 hrs, 7 days and 30 days after operations tespectively.6 ml of 25% Sodium fluorescein were given intravenously,30 minutes before the animals were sacrificed, the fluorescein-infarcted area existed on the slices in different size and degree in the territory which supplied by the right MCA. Though it was considered that extravasation of the sodium fluores cein from the cerebral vessels were due to broken down of the brain-blood-barrier and severe acute cerebral ischemic lesions.The model of cerebral infarction may be duplicated by this method and the infarcted rate was more higher, according to our data the rate of infarction was 100%, we think it will provides great benefit for experimental therapeutic research of acute cerebral infarction in the future.

1984年以来我们采用14只杂种狗进行大脑中动脉区脑梗塞模型建立的实验性研究。开颅,将右侧ICA远端及MCA近端同时夹闭后,每日按Crowell及Olsson5级法进行临床观察和记录。按计划将求后存活的狗分别于术后24小时、7天和30天处死,其中10只术后7天处死,均发现在右侧MCA区有大小和程度不等的荧光——梗塞区存在,考虑系由严重的脑缺血损害和血脑屏障的崩解引起荧光素钠外渗和滞留的结果。本实验方法可以复制而且具有较高的梗塞率,本实验组梗塞率达到100%,对今后脑梗塞急性期实验性治疗的研究,提供了良好的条件。

The purpose of this experiment was to study the role of arginine vasopressin (AW) in acute cerebral ischemic edema in mongolian gerbils. The results showed that intracerebroventricular injection (ICV) of AVP exacerbated the ischemic brain edema, while ICV of AW antiserum significantly decreased the ischemic brain edema. Nimodipine couldn't block this role of AW in ischemic brain edema. The cortical Na+ -K+ ATPase activity was significantly decreased, the contents of cAMP in the ischemic...

The purpose of this experiment was to study the role of arginine vasopressin (AW) in acute cerebral ischemic edema in mongolian gerbils. Nimodipine couldn't block this role of AW in ischemic brain edema. The cortical Na+ -K+ ATPase activity was significantly decreased, the contents of cAMP in the ischemic cortex and hypothalamus and the contents of cGMP in the hypothalamus were remarkably increased after ICV of AW. These suggest AW was involved in the pathophysiologic process of acute ischemic brain edema. And its mechanism might be the effect of AW on AW receptor mediated by cAMP, cGMP, and that in turn inhibited the Na+ -K+ ATPase activity of brain cell membrane, then exaggerated the formation of ischemic brain edema.

本实验探讨了精氨酸加压素(AVP)对蒙古种沙土鼠急性缺血性脑水肿的作用及其机理。结果表明,给沙土鼠侧脑室注射AVP后,缺血性皮质水肿显著加重,尼莫地平不能阻断AVP对缺血性脑水肿的影响;侧脑室注射AVP抗血清后,缺血性皮质水肿明显减轻,侧脑室注射AVP后,缺血性皮质Na~+-K~+ATP酶活力显著降低,皮质和下丘脑cAMP以及下丘脑cGMP含量均显著升高。提示AVP参与了急性缺血性脑水肿的病理生理过程,且起着促进或加重缺血性脑水肿形成的作用。其作用机理可能是通过cAMP、cGMP介导的AVP受体的作用,产生抑制脑细胞膜Na~+-K~+ATP酶活力的效应,从而促进缺血性脑水肿的形成。
