This is a study of the fine structure of chloroplast in the cytoplasmic male- sterile line (chi) Isuzn and its maintainer Isuzu in oil rape.Both experimental materials were gifts by Japanese Institute of Agriculture Techniques in 1980.Seeds of rape were planted early spring in 1981 in the green house under the experimental station of the Institute and bloomed in May through natural vernalization.Leaf blade bearing on the top of plant was fixed before blooming in 4% phosphate-buffered ghitaraldehyde at 4C for 2 hr and rinsed with buffer solution,then postfixed 4 hr in 1 % osmium tetroxide at 4C and rinsed with double distilled water.After dehydration in a graded series of ethyl alcohol and propylene oxide,the samples were then embedded in Epon 812 and sectioned on an LKB-4800 ultramicrotome.Sections were stained with ur-anyl acetate and lead citrate and examined and photographed with JEM-CX electron microscope.In the chloroplast of maintainer the structure of thylakoid lamellar system is normal and composed of both grana and stroma lamellae.The whole lamellar system is well arranged and organized in order.There are about 10 grana in each chloroplast and 4-10 lisks in each granum.This feature of chloroplast structure is similar to that of other higher plant species.But there is another picture of chloroplast structure in the cytoplasmic male-sterile line.The lamellar system involves elongated thylakoid lamellae.The grana are composed of 2-4 discs of thylakoid lamellae and are elongated in form also.Some thylakoid lamellae connecting the elongated grana are broken up,therefore,the arrangement of the lamellar system seems to be disordered.Similar features were observed by others in the chloroplast mutants in maize,barley and Chlamydomonas.

Six gene libraries of Saccharomyces cerevisiae 7209-lA(a) were constructed efficiently with cosmid pHC79 DNA ligated at their BamHI,PstI,and HindIII sites,using HB101 and SC294 as recipient strains.Several factors affected the construction of the libraries.With proper extent of the restriction enzyme digestion of yeast DNA,F/V ratio for ligation at 15/1,DNA concentration for ligation at 200-250 ng/ml and BHB2688/BHB2690 for in vitro packaging at 5/1,we got rather high efficiency of library construction (106 CFU/μg vector DNA) and low Apr Tcr % (down to less than 10% or even 5%).We found the complementation (supression) effect on the auxotrophs of leuB,proA (in HB101),Ieu6,metB1,his1,and argG6 (in SC294,a gift from Dr.G.Hobom).The frequencies of complementation of these six genes are between 10-3-10-4,while the spontaneous reversion rates in leu,pro,his,met,and arg mutants are lower than 10-4.As a result,we conclude that genes (Leu,Pro,Met,His,and Arg) on the inserted fragment of yeast DNA in the hybrid plasmid can be functionally expressed in E.coli.We also found high frequency (>30%) of cotransduction of Pro+with Leu+pheno-type in yeast gene library in HB101.We have concentrated yeast Leu2 gene or supres-sor for more than 104 times after repackaging and retesting for complementation witb pYeleu5 and pYeleu7 DNA.

Tangerine (Citras reticutata var.tanoerina Tan.)is a lind of wounderful fruit as edibles, gift and ornament during the Spring Festival in China. It will satisfy the needs of the festival and export if the fruits Are retained with leaves and preserved fresh. It has been proved in this experiment that the fruits can be preserved fresh and retained leaves for 48 days after soaked with the No. 1 mixed fruit preserving agent. The fruits with leaves loaded in bamboo basket in batches and transported 160 km. Still retained their leaves 65-88% for 12 days, and weight loss being 1.19% only. These fruit still looked plump, lustrous, and in good color with green leaves.Sale trails were sought after by customers. The commercialized experiment should be carried out on a large scale in the near future. Experiment with labelled compounds of 32p-NaHPO4, 3H-glucose, 3H-H3O and 14C-2,4-D indicated that the action of exogenous hormone and moisture is the key factor on preserving fresh and retaining leaves of tangerine. Routine analysis carried Out in the preserved fruit revealed that no evident loss of nutrients occurred. The residue of chemicals had been checked by gas chromatography, and the content of batistin was within the limits of international standard, with the exception of one sample treated with 50 Or 200 ppm of 2,4-D, of with showed a little higher than the international standard; but all other samples were lower than the international standard. So it is safe for edibleness.

本试验证明采用1号复合保鲜剂能使年桔[Gitrus reticulata var.(langerina)Tan.]有效地带叶保鲜,桔红叶绿,更能满足春节期间送礼、观赏和食用的需要,即使落叶后也能延长贮存期。经四年较大批量的保鲜运输、贮存和试销获得好评。利用示踪原子法研究结果表明,年桔带叶保鲜的关键因子是外源激素和水分的作用。
