Proliferative survival curves of Eca109 cells treated with hematoporphyrin derivatives (HpD) and light varied with the dosage of HpD and duration of light exposure. The survival curve of cells treated with 1μg/ml HpD and 2~10 minutes irradiation was similar to that of the control. When the dosage of HpD was increased to 2,3 and 5μg/ml, cloning efficiencies of cancer cells decreased as the exposure duration prolonged. Autoradiography showed slight decrease in incoporating activities of 3H-TdR, 3H-UrD and 3H-tyrosine in treated cells and recovery from these injuries 2~5 days later. By Coomassie blue staining, network structure appeared in cells following photosensitization instead of filamentous organelle. Filamentous cytoskeleton reformed 48 hours after photosensitization. Five kinds of enzymes stained by histochemical methods showed similar results in Eca109 and LTEP-78 cells:i.e., 1) activity of succinic dehydro-genase in mitochondria and acid phosphatase in lysosomes decreased instantly after treatment and increased again 1-6 days later, 2) monoamine oxidase increased slightly and 3) alkaline phosphatase and 5′ -nucleotidase showed no difference between treated and control cells. The most serious injury in protein of filamentous organelle and enzyme in mitochondria occurred 6 hours to one day after treatment. Cells damaged slightly in structure or in function could recover from the damage. The mechanism of photodynamic injury was discussed.
