Western blot, immunoprecipitation, membrane cross-linking and PTK activity were developed with anti-fyn, anti-phosphotyrosine and anti-p subunit of GM-CSF receptor antibodies. In murine bone marrow derived macrophages (BMDM) GM-CSF stimulation could lead to several cyto-plasmic proteins tyrosine phosphorylation within 15 minutes. Among them phosphoprotein p59 was revealed to be fyn oncoprotein. Simultaneously, fyn PTK activity was enhanced over two-fold. Membrane cross-linking displayed that a membrane complex (p330 450) was formed after GM-CSF stimulation in which p59 fyn might link to P subunit of GM-CSF receptors.

本实验研究了小鼠巨噬细胞中原癌基因fyn酪氨酸蛋白激酶(PTK)的活性和酪氨酸磷酸化过程在粒-巨噬细胞集落刺激因子(GM-CSF)受体诱导的信号转导中的作用。用抗fyn,抗磷酸化酪氨酸和抗GM-CSF受体(β链)单抗进行了蛋白免疫印迹、免疫沉淀、膜交联以及PTK活性测定。结果发现在小鼠骨髓来源的巨噬细胞(BMDM)中,GM-CSF刺激后15分钟内,可见细胞浆中数种蛋白的酪氨酸磷酸化,其中p59被证实是fyn癌基因蛋白。同时,fyn酪氨酸蛋白激酶(PTK)活性 在GM-CSF刺激后被增强二倍以上。细胞膜交联实验显示在GM-CSF刺激后形成的p330-450膜复合体中p59fyn蛋白与GM-CSF受体β链相关联。
