The propagation properties of orf virut in primary dairy-caprine testis cell culture were sfudied. Three strains of orf virus, HCV. LTV and AGV Were isolated from the scabs of oral lesiolls of goats in Huang-Cheng, Ling-Tai and A-Yan-Gou pastures. A comparative study on the adsorption passage, mixing passage and modified passa- ge method were carried out. To reveal the relationships between adsorption time and CPE, viral concentration and CPE on the one band, and those among viral replication, CPE and incubation time on the other, the orf virus adapted to cell culture, the 15th passage HCV strain was used. The physical-chemical properties of the strain adapted to cell culture were compared with those of the field HCV strain. Experimental results showed that at lst-10th bpassa CgPE appeared irregularly and had a significantd ifference etween eache strain but after the 1oth passage, CPE tended to be stable. At 42 hours after infection, the replicatipn of the virus reached up to ifs peak, infectivity titre was 10~6 TCID50/0. 2ml. about 50 to 60% cells showed specific CPE. The virions budding from cell membranes have additional membranes. Lt was found that there were twoor thre e capsids in some virions.

本文采用奶山羊睾丸细胞对羊口疮病毒的繁殖特性进行了研究。从来自皇城、灵台和阿沿沟牧场的病羊痂皮组织中分到HC、LT和AT三株口疮病毒。比较了吸附一、混合一、改进混合三种方法分离培养口疮病毒的效果。进一步选用第15代HC分离物分别测定了吸附时间、接种浓度同细胞致病作用(CPE)、以及病毒复制、CPE与培养时间的关系,对比了细胞毒与痂皮毒的理化特性。结果表明,1—10代CPE很不规则,且有显著的毒株差异,而10代以后CPE趋于稳定。病毒复制于感染后42小时达到高峰,效价为10~6TCID_50/0.2ml,50—60%CPE为其标指;芽生释放的病毒具有一层外膜结构。还发现具有2或3个核衣壳的病毒粒子。

目的  根据登革病毒 (DEN) 3 端非编码区的一段高度保守序列 ,设计登革 1~ 4型通用引物 ,采用逆转录聚合酶链反应 (RT PCR)检测登革病毒 1~ 4型RNA。方法 用C6/ 3 6细胞培养登革病毒。用热酚法提取病毒RNA ,进行RT PCR扩增 ,并对扩增产物测序。结果 DEN 2 NGC株可扩增出至少 10TCID5 0 的病毒 ,测序结果与已知序列一致。DEN 1~ 4型标准株和DEN -2 -0 4与DEN 2 4 3株均能够扩增出唯一的一条特异条带。结论 应用通用引物RT PCR法可从病毒浓度少至 10TCID5 0 的感染细胞培养液中检出DEN 2 NGC株病毒 ,并且该对通用引物对于其他 3型登革病毒亦具有很好的通用性 ,其方法的特异性和敏感性亦较强。
