Allergic asthma caused by Pleurotus sapidus besidiospores occurs in high incidence and brings about great hazard among mushroom cultivating workers in China.In order to elucidate the role of cytokines in the pathogenesis of this disease,we set up an animal model of allergic asthma through the challenge of Pleurotus sapidus besidiospores in primed rats,and measured a series of related cytokines and their receptor mRNA levels of spleen lymphocytes from rats with asthma.SDS PAGE analysis showed that the besidiospore antigen is a 17kDa protein.Cytokines and their receptor expression at mRNA levels were determined by RNA/cDNA hybridization using radiolabelled cDNA probes for different cytokines and their receptors in vitro. Results showed that mRNA expression of IL 4,GM CSF,IL 6,IL 2 and IL 2R,but IL 6R in lymphocytes of asthma suffering rats challenged for 7d increased by 54%,45%,170%,83% and 76%,respectively.In rats challenged for 2d,mRNA levels of IL 4,IL 6 and IL 2 increased respectively by 37%,58% and 125%;whereas mRNA level of GM CSF,IL 2R and IL 6R unchanged.Results suggest that allergic asthma induced by Pleurotus sapidus besidiospores markedly activates IL 4,GM CSF,IL 6,IL 2 and IL 2R,except IL 6R expression in lymphocytes as evaluated at mRNA level.We consider that there exists a significantly increase in T H2 type cytokines in the pathogenesis of Pleurotus sapidus besidiospore induced asthma in which IL 4 may play an essential role.

CD4+ helper T lymphocytes and γδ T cells play important role in antimicrobial immunity, however, the mechanism of interactions in question still remains to be elucidated. Methods CD4+T cells and γδ T cells separated from peripheral blood lymphocytes of healthy donors by panning technique were stimulated with intracellular pathogen-Mycobacterium tuberculosis and human herpes virus-6 as well as extracellular pathogen-TrichomonasVaginalis respectively in vitro. Results The stimuli induced CD4+ T cells significantly to proliferate and producing Th1-type cytokine profile-IL-2 and IFN-γ. With a semipermeable co-culture system in vitro, antigen-activated CD4+ Th1-like cells were found to promote the proliferation and cytotoxicity of γδ T cells via soluble factors. Conclusion Functions of γδ T cells might be inhanced by CD4+ cells with Th1-type cytokines in antimicrobia1 immunity.

CD4+T辅助(Th)细胞和γδT细胞在抗微生物感染中发挥着重要作用,两者相互作用有待阐明。本文报道以胞内感染体结核杆菌、人类VI型疱疹病毒及胞外寄生虫阴道毛滴虫刺激活化从正常人外周血经洗淘法分离纯化的CD4+T细胞,结果各组活化的细胞均表现明显增殖和产生以白介素-2(IL-2)、γ干扰素(IFN-γ)显著升高为主的Th1型细胞因子分泌谱。利用细胞隔离渗透性共培养体系,将抗原活化的CD4+T细胞和γδT细胞共培养,结果显示γδT细胞的增殖和细胞毒作用均可为CD4+T细胞产生的可溶性因子所显著增强。这提示在抗感染免疫中,CD4+Th1细胞可通过Th1型细胞因子促进γδT细胞功能。

Objective In order to investigate the function of γδ T cells,this study has been focused on the cytotoxicity and cytokine production of pathogen activated γδ T cells as well as their influence on the proliferation of CD4 + cells.Methods γδ T cells were separated from peripheral blood of healthy donors by panning.After stimulated with PHA, Mycobacterium tuberculosis (M.tb),human herpes virus 6(HHV 6) and Trichomonas Vaginalis (TV),γδ T cells were detected for their cytotoxicity and cytokine production,as well as effect of γδ T cell derived soluble factors on the growth of CD4 + cells in a semi permeatable membrane co culture system. Results M.tb could induce higher cytolytic activity of γδ T cells to K562 cell line than TV.M.tb and HHV 6 could stimulate γδ T cells predominantely to produce IFN γ,while TV could trigger γδ T cells mainly to secrete IL 6.The soluble factors secreted by pathogen stimulated γδ T cells could promote the proliferation of CD4 + cells. Conclusions Intracellular pathogens can be induciable factors for the production of T H1 type cytokines by γδ T cells and promote the cytotoxicity of γδ T cells,whereas the extracellular pathogen can induce γδ T cells to produce T H2 type cytokines.

目的体外研究病原体抗原活化的γδT细胞的细胞毒和细胞因子产生的反应性以及对CD4+T细胞增殖的影响,以期进一步澄清γδT细胞的功能。方法用洗淘法从外周血中分离纯化出γδT细胞,在体外探讨植物血凝素和结核分枝杆菌(M.tb)、人类6型疱疹病毒(HHV-6)和阴道毛滴虫(TV)病原体抗原对γδT细胞的细胞毒活性和细胞因子产生的影响,并采用细胞隔离渗透培养法观察γδT细胞分泌的细胞因子对CD4+T细胞增殖的作用。结果M.tb比TV可更强地促进γδT细胞对K562细胞的杀伤活性。M.tb和HHV-6可刺激γδT细胞主要分泌TH1型细胞因子IFN-γ,而TV可刺激γδT细胞优势产生TH2型细胞因子IL-6。经抗原活化的γδT细胞分泌的可溶性因子可促进CD4+T细胞增殖。结论细胞内感染的病原体是激发γδT细胞产生TH1型细胞因子的诱导因子,并对其杀伤活性有所增强,而细胞外感染病原体可激发γδT细胞产生TH2型的细胞因子。
