The effects of sophoridine (Sop, 30 mg / kg iv) on ischemic ventricular tachyarrhythmias and electrophysiologic consequences in both normal and ischemic myocardium were studied in the open+chest dogs subject to programmed electrical stimulation (PES) oh 5-8 d after acute myocardial infarction. The effects of sophoridine (Sop, 30 mg / kg iv) on ischemic ventricular tachyarrhythmias and electrophysiologic consequences in both normal and ischemic myocardium were studied in the open+chest dogs subject to programmed electrical stimulation (PES) oh 5-8 d after acute myocardial infarction. Its effects were compared with those of procainamide (PA 25 mg / kg iv). Both drugs distinctly lengthened the QTc interval and the effective refractory period (ERP) of the normal and infarct myocardium in both ventricles, decreased the dispersion of ERP in infarct myocardium (IDR) as well as the dispersion of ERP in left ventricle (VDR) and increased the diastolic excitability threshold(DET) of the normal and infarct myocardium in both ventricles remarkably. The PES-induced ventricular tachycardia (VT) or ventricular fibrillation (VF) was prevented in the 5 out of the 6 Sop and PA treated dogs respectively. Sop prevented spontaneous VF in 5 dogs (n = 6); PA prevented spontaneous VF in 4 dogs (n = 6); normal saline (NS) did not prevented PES-induced VT or VF and spontaneous VF. The results suggested that Sop might be effective in preventing the onset of reentrant ventricular tachyarrhythmias and sudden coronary death after myocardial ischemic damage.

The effect of AMG- 1 [6( 5 - hydroxy- 2- methylpyridylamino ) - 9 ribofranosylpurine] on mouse brain energy metabolism in complete brain ischemia induced by decapitation and on neurological deficit and histopathological changes after occlusion of the middle cerebral artery (MCAO) in rats were studied. The results indicate that AMG-1 has an effect of improving the status of energy metabolism in complete brain ischemia in mice. Lactate concentration was greatly reduced and the ATP and phosphocreatine levels were significantly elevated. Treatment with AMG-1 or nimodipine was performed before and after MCAO. The score of neurological deficit was significantly decreased as compared with the vehicle treated group. The extent of ischemic neuronal injury was determined by histopathological examination. AMG- 1 and nimodipine seemed to attenuate the MCAO - induced neuronal damage. In as much as AMG- 1 can improve the status of brain energy metabolism, neurological deficit and neuronal damage after ischemic insult, AMG- 1 may have a beneficial effect for the treatment of cerebral ischemic damage.

本文观察AMG-1对小鼠断头全脑缺血时能量代谢及大鼠大脑中动脉阻断(MCAO)后行为和病理改变影响。小鼠sc AMG-1 1~10mg/kg 30min后,能有效减少脑缺血后乳酸(LA)堆积,ATP和磷酸肌酸(PCr)的耗竭。sc AMG-1 5mg/kg和尼英地平0.5 mg/kg,可减轻大鼠MCAO后的神经症状和神经细胞缺血性损害。AMG-1的这一改善脑缺血作用与尼莫地平近似。
