AIM: To investigate the effect of the body moderate hypothermia inhibition on apoptosis after cerebral ischemia and reperfusion injury and discuss the mechanism of hypothermia on cerebral resuscitation and the stimulative factors of postischemic apotosis. METHODS: Using the globalischemia and reperfusion model in rat according to the modified procedure of Pulsinelli et al, changes of neuron apoptosis in hippocampus were detected by a TdTmediated dUTPbiotin nick end labeling (TUNEL) method. RESULTS: A few blue grains were observed in some cells in hippocampal CA1 with a 6hour reperfusion performed 20 minutes after the cerebral ischemia, and the blue cells increased markedly 12 hours, 24 hours, 48 hours, 3 days and 5 days after the reperfusion. Apoptotic cells peaked on the 5th day and began to drop from the 7th day. Treatment with body moderate hypothermia significantly reduced the number of apoptotic cells. CONCLUSION: ZThe body moderate hypothemic of cerebral resuscitation can inhibit or delay postischemic apoptosis by inhibiting or blocking the cascade reaction of apoptosis.

目的:观察全身亚低温对大鼠全脑缺血再灌注损伤细胞凋亡的影响,探讨亚低温的脑复苏机制和脑缺血后凋亡刺激因素.方法:采用改良的Pulsineli-WA4VO法,制备大鼠全脑缺血再灌注模型,采用TUNEL法,观察海马脑区细胞凋亡的特征变化.结果:发现脑缺血20min再灌注6h,海马CA1区可见少量细胞核呈蓝染颗粒,于再灌注12h,24h,48h,3d,5d蓝染阳性细胞明显增多,5d达高峰,7d开始下降.给予亚低温治疗后上述变化明显减轻.结论:亚低温脑复苏作用可能是通过抑制或延迟脑缺血后细胞凋亡,而抑制或阻断凋亡发生的启动级联反应为其重要机制.

AIM: To observe the effect and mechanism of whole body moderate hypothermia on the expression of related gene after global cerebral ischemia and reperfusion. METHODS: According to the modified procedure of pulsinellis 4-vessel occlusion, we observed the changes of expression of bcl-2 and bax in ischemia group and the influence of hypothermia on these changes using immunohistochemistry assay. RESULTS: The level of bcl-2 expression was low at 6 h post-ischemia injury and it increased gradually with the passage of time. The expression reached peak level at 24 h and reduced at 7 d. Low expression was also observed in hypothermia group but it reached the peak at 3 d. Significant difference in the expression level of bcl-2 at 3 d was found between ischemia group and hypothermia group (P<0.01). The level of bax expression increased at 6 h after ischemia, reached the peak at 5 d and remained high at 7 d. Compared with that in ischemia group, the expression of bax were markedly lower at 5 d and 7 d (P< 0.01) in hypothermia group and no peak was observed. CONCLUSION: Hypothermia can inhibit the expression of bax, the gene that promotes neuronal apoptosis. The inhibition of neuronal apoptosis may contribute to the preventive effect of hypothermia on global ischemia.
