The retinoblastoma susceptibility gene(Rb) was the firstly identified tumor suppressor gene. But the expression of Rb and its product. PRb, in oral carcinogenesis has not been reported. It was found immunohistochemically that the positive reactions of PRboccurred in cellular nuclei and cytoplasms of tissues in each stage of oral carcinogenesis.The positive staining in nuclei was stronger than that in cytoplasms and positive patterns showed heterogenecity.It seemed that PRb expression associaseiated with the differential degrees of invasive squamous cell carcinomas. These results support to some extent our speculations from the research on P16 expression in oral carcinogenesis and indicate that it is necessary to study further the changes and effects of Rb gene and its product PRb on the development of oral carciongenesis.

AIM To discuss the expression and correlation of retinoblastoma susceptibility gene and proliferating cell nu clear antigen in the prostatic cancer. METHODS SABC immunohistochemistry was used to determinate Rb gene and PCNA in 36 samples of the prostatic cancer. The image pattern analyzer and the statistics methods were used to do the comparative analysis of Rb gene and PCNA in the different pathological grading and clinical phases of the prostatic cancer. RESULTS immunohistochemistry staining was Rb positive in 17 of 36 samples,rated 47.2%. The expression of Rb had significant differences in different pathological grading and clinical phases ( P <0.05). The expression of PCNA also had significant differences in different pathological grading and clinical phases ( P <0.05). The expression of PCNA was weak in the Rb positive expression samples and was strong in the Rb negative expression samples. There was a significant difference between Rb and PCNA expression ( P <0.05). CONCLUSION Rb is involved in the occurrence and development of the prostatic cancer. As for pathological mechanism, it is related to the abnormality of Rb and PCNA regulating path. The determination of Rb and PCNA can be an index for estimating the level of malignant and progression of the prostatic cancer.

目的 探讨视网膜母细胞瘤易感基因 (Rb)和增殖细胞核抗原 (PCNA)在前列腺癌中表达的意义及其相互关系 .方法 采用免疫组织化学 SABC法对 36例前列腺癌标本 Rb基因和 PCNA进行检测 ,并应用图像分析仪及统计学方法对两者在不同病理分级及临床分期中的表达进行对比及相关分析 .结果  36例前列腺癌标本中 Rb表达阳性 17例 ,阳性率47.2 % ,其表达在前列腺癌的不同病理分级及临床分期之间有显著差异 (P <0 .0 5 ) ;PCNA在标本中的表达强度在不同病理分级及临床分期之间有显著差异 (P<0 .0 5 ) ;同时还发现 Rb表达与 PCNA表达存在负相关 ,即 Rb阳性标本中 PC-NA表达弱而 Rb阴性标本中 PCNA表达强 ,两者比较有显著性差异 (P<0 .0 5 ) .结论  Rb基因参与了前列腺癌的发生、发展过程 ,在发病机制中可能涉及到 Rb和 PCNA调节通路的异常 ,检测 Rb基因和 PCNA有助于前列腺癌恶性程度的判定及预后 .

Objective: To study the expression of retinoblastoma susceptibility gene(Rb) and minichromosome maintenance protein 2(MCM2) in primary prostate cancer(PCa) and its clinical significance.Methods: The expression of MCM2 protein and pRb were detected in 49 cases of PCa,20 cases of benign prostate hyperplasia(BPH),10 cases of normal prostate tissues(NP) by EliVisionTM plus immunohistochemical staining.Results: The positive expression rate of pRb in PCa,BPH,NP were 44.90%,80% and 90% respectively.The expression level of pRb in PCa was significantly lower than that in BPH(P<0.05) and NP(P<0.05),and was negatively correlated with pathological grades(P<0.05) and clinical stages(P<0.05) of PCa.The positive expression rate of MCM2 in PCa,BPH,NP were 67.35%,25% and 10% respectively,however,the expression level of MCM2 in PCa was significantly higher than that in BPH(P<0.05) and NP(P<0.05),and was positively correlated with pathological grades(P<0.05) and clinical stages(P<0.05) of PCa.The expression level of MCM2 in PCa was negatively correlated with pRb(rs =-0.596,P <0.01).Conclusion: Rb might be involved in the occurrence and development of PCa.The determination of Rb and MCM2 can be an index for estimating the level of malignancy and prognosis of PCa.

目的:探讨视网膜母细胞瘤易感基因(Rb)和微染色体维持蛋白2(MCM2)在前列腺癌发病机制中的作用及其临床意义。方法:应用免疫组化EliVisionTM plus二步法,检测49例前列腺癌(PCa)组织,20例良性前列腺增生(BPH)及10例正常前列腺组织(NP)中pRb蛋白与M CM 2的表达。结果:pRb在PCa,BPH与NP组织中的阳性表达率分别为44.90%,80%,90%,PCa中pRb的水平明显低于BPH(P<0.05)及NP(P<0.05),且与PCa病理分级(P<0.05)和临床分期(P<0.05)呈负相关。M CM 2在PCa,BPH与NP组织中的阳性表达率分别为67.35%,25%和10%,PCa中M CM 2的水平明显高于BPH(P<0.05)及NP(P<0.05),且与PCa病理分级(P<0.05)和临床分期(P<0.05)呈正相关。M CM 2表达与pRb表达呈负相关(rs=-0.596,P<0.01)。结论:Rb基因参与了前列腺癌的发生、发展过程,检测Rb基因和M CM 2有助于判定前列腺癌恶性程度及预后。
