Objective To investigate the roles of salvia miltiorrhiza monomer IH764-3 in anti-fibrogenesis and the mechanism of collagen degradation.Methods Forty male Sprague-Dawley rats.were randomly divided into three groups,named control group, model group and IH764-3 treatment group.The rats of treatment group received 40 mg·kg-1 of IH764-3 intraperitoneally , while other rats were injected intraperitoneally by equivalent saline. The tissue sections were stained immunohis-tochemistrically to detect TIMP-1 and TGF-β1, and positive areas were measured. Results The secondary biliary hepatic fibrosis was induced successfully. Treated by IH764-3 , the content of hydroxyproline decreased by 40.6% . Immunohistochemistrically, the positive areas of TIMP-1 and TGF-β1 decreased by 36.2% and 32.5% , respectively. Conclusions Salvia mitiorrhiza monomer IH764-3 may relieve hepatic fibrosis induced by bile duct ligation. One of the mechanism of this effect seems to be i n relation to decrease expression of TIMP-1 through inhibiting expression of TGF-β1.

目的 研究丹参单体IH764-3抗胆总管结扎大鼠肝纤维化作用,观察其对胶原降解的影响。方法 40只大鼠随机分为对照组、模型组及IH764-3预防组。预防组腹腔注射IH764-3 40mg·kg~(-1)·d~(-1),另两组注射等剂量生理盐水,共14天。应用免疫组织化学方法检测TIMP-1、TGF-β_1。结果 胆总管结扎大鼠14天后有明显肝纤维化形成,IH764-3可使肝组织内羟脯氨酸减少40.6%,同时TIMP-1、TGF-β_1蛋白质表达分别下降36.2%和32.5%。结论 丹参单体IH764-3可以减轻胆总管结扎大鼠肝纤维化程度,其抗肝纤维化作用机制之一是抑制TIMP-1和TGF-β_1蛋白的表达。

To observe the role of salvia miltiorrhiza monomer IH764-3 in anti-fibrogenesis in vivo and in vitro, especially the mechanism involved in collagen degradation. Methods: Forty male SD rats were divided into three groups at random, namely control group(n=10), model group (n=15) and IH764-3 group(n=15). The experimental hepatic fibrosis was induced by common bile duct ligation(BDL). The tissue sections and the cover slides of hepatic stellate cells(HSC) were stained with immunohistochemistry to detect tissue inhibitor of matrix metalloproteinase-1(TIMP-1), transforming growth factor beta-1 (TGF-β_1) and hydroxyproline(Hyp) positive areas of tissue sections were measured. Results: Obvious hepatic fibrosis had come into being 14 days after BDL. IH764-3 decreased the content of Hyp by 40.6% and the positive area of TIMP-1 and TGF-β_1 by 36.2% and 32.5% respectively. In groups of HSCs exposed to different dose of IH764-3, the numbers of positive cells of TIMP-1 and TGF-β_1 decreased evidently. Conclusion: Silvia mitiorrhiza monomer IH764-3 may relieve hepatic fibrosis in vivo and in vitro, one of the mechanisms of which seems to be relative to decreasing expression of TIMP-1 through inhibiting expression of TGF-β_1.

目的:研究丹参单体IH764-3在大鼠体内、外的抗肝纤维化作用,重点观察对胶原降解的影响。方法:40只SD大鼠随机分为3组,对照组10只,模型组及IH764-3组各15只。以胆总管结扎法(BDL)建立肝纤维化模型,用免疫组化法检测大鼠组织血细胞金属蛋白酶组织抑制因子(TIMP-1)、转化生长因子β_1(TGF-β_1)和肝组织羟脯氨酸(Hyp)含量。结果:BDL结扎14d后有明显肝纤维化形成。IH764-3使肝组织Hyp减少40.6%,同时TIMP-1、TGF-β_1蛋白质表达分别下降36.2%和32.5%;在体外使TIMP-1、TGF-β_1阳性肝星状细胞数目减少。结论:丹参单体IH764-3具有抗肝纤维化作用,其机制之一是抑制TIMP-1和TGF-β_1蛋白的表达。

Objective To evaluate the relationship between serum testosterone (T) and anti-fibrogenesis of 17β-estradiol (β-Est). Methods Liver fibrosis was induced by CCl4 administration. The fibrosis-suppressive effect of β-Est (20 μg·kg-1·d-1 ) was evaluated in the intact or gonadectomized male rat model. The serum levels of liver enzyme, extracelluar matrix (ECM) and sex hormone were determined by standard enzymatic methods, ELISA and RIA, respectively; the degrees of fibrosis in the liver were determined by VG stain; transforming growth factor-β1 (TGF-β1 ) and activin βA( ACTβA) expressions in the liver were studied by immunohistochemistry and semiquantita-tive RT-PCR. Results All the changes made it clear that β-Est treatment could improve the serum levels of liver enzyme, reduce ECM secretion, suppress hepatic collagen deposition and decrease TGF-β1 and ACTβA expression in the gonadectomized male rat model significantly, but not in the intact male rat model despite attaining the same serum estradiol ( E2 ) levels. Conclusion β-Est could reduce CCl4-induced hepatic fibrosis in rats, but the resistance to the protective effects of estrogen in the male appears to depend on the presence of testosterone. This may be one reason for the sex associated differences in the progression from hepatic fibrosis to cirrhosis.

目的探讨体内睾酮(T)和雌激素(β-Est)抑制肝纤维化的关系。方法CCl4皮下注射诱导肝纤维化模型,观察20μg·kg-1·d-1β-Est对完整或去势雄性大鼠肝纤维化形成的影响。以标准酶、ELISA、RIA分别测定血清肝功能、细胞外基质(ECM)和性激素水平;VG胶原染色观察肝组织病理学改变,结合图像分析计算胶原面积;RT-PCR和免疫组化检测转化生长因子-β1(TGF-β1)、激活素BA(ACTβA)在组织中的表达。结果β-Est能改善去势纤维化大鼠肝功能,降低ECM分泌、肝组织纤维化程度和TGF-β1、ACTβA表达,而对完整雄性大鼠肝纤维化形成无影响;两组大鼠血清雌二醇水平无显著性差异。结论β-Est能抑制肝纤维化形成,T影响其发挥保护作用,可能是造成肝纤维化性别差异原因之一。
