The results defined that with H. E staining,the spores were found to be charac teristically surrounded by a narrow clear halo. The lectin binding pattern of the organisms in tissue sec tions was similar to that in culture,which was different from several other kinds of deep fungi. It was found that the semi-thin section embedded in Epon had an outstanding advantage both in detecting the organisms and showing the relation between the organisms and the host cell. The results showed that the morphologic changes of the organisms in tissue related to the host resistance.

Objective In order to explore a new method of aetiological diagnosis for deep fungal infection in vivo.Methods The lectin binding patterns of 17 deep fungi (include specimens from cultures and tissue samples from experimentally infected mice)were studied,using 14 different biotinyl lectins and avidin-horseradish peroxidase(ABC).Results Lectin binding patterns of the tested fungi were stable.Most of the tested fungi were distinguishable each other based on the lectin binding patterns of the genus,few were distinguishable of the species.Conclusion The mentioned results offer a experimental basis for differentiating fungi in tissue section using the lectin histochemistry.

目的探索深部真菌病组织内病原菌检查的新方法。方法应用生物素卵白素过氧化酶法(ABC)研究17种深部真菌(包括培养中真菌及小鼠实验性感染皮肤及内脏组织内真菌)与14种凝集素的结合形式。结果17种真菌的凝集素结合形式稳定。绝大多数真菌的凝集素结合形式呈属间区别,少数亦见种间区别。结论为利用凝集素组织化学鉴定组织内病原菌提供了实验依据。

Objective To study the lectin binding patterns of fungi in histological sections and in smears of sodium hydroxide lysed nail from the patients with onychomycosis. Methods Using 12 biotinyl lectins and avidin horseradish peroxidase(ABC method), the binding patterns of the fungi in the histological sections and nail lysates from 72 cases of onychomycosis were studied and compared with their counterparts in the published reports. Results All the lectin binding patterns of 7 fungi in the histological sections and nail lysates from onychomycosis patients showed identical with their counterparts in the culture, except that Microsporum ferrugineum stained positively with pisum sativum agglutinin in tissues while negatively in the culture. The nail lysate method revealed advantages of high positive rate and time saving procedure. Conclusion The lectin binding patterns of the fungi in histological sections and nail lysates appear identical with their cultured counterparts and may be helpful in mycological identification in causal fungi in onychomycosis.

目的 探讨甲真菌病组织切片、溶甲涂片中真菌的凝集素结合形式,并检测其与培养中相应真菌凝集素结合形式是否等同。 方法 对已分离鉴定致病菌的 72例甲真菌病的病甲组织切片、溶甲涂片,用 ABC法对 12种凝集素进行组化染色,并与已知培养中的相应菌种进行比较。结果 44例甲组织切片、 28例溶甲涂片内 7个种、属的真菌与其培养中的相应真菌呈现相同的凝集素结合形式,仅铁锈色小孢子菌豌豆凝集素的结合形式与培养中不同,从而表明真菌表面的糖蛋白作为凝集素受体的结构颇为稳定,在经过标本制备处理后仍保持种、属的特点。溶甲涂片方法还具有阳性率高和省时的优点。结论甲组织切片、溶甲涂片内真菌的凝集素结合形式与培养中相应真菌具有相同的凝集素结合形式,有助于甲真菌病病原菌的鉴定。
