A method of the ion pair extractiveseparation and subsequent spectrophotome-tric determination of aconitine and aconitealkaloids in the preparation is reported. The method is based upon the forma-tion of ion pair addition compound ofalkaloids and bromothymol blue in bufferedsolution (pH6.0). The ion pair additioncompound was extracted with chloroformand the absorbance of the solution wasmeasured at 410nm against a reagent blank.The absorption curve is linear in the rangeof 65-259μg of aconitine. The recovery ofthe hydrolysis products of aconitine is 97.64±1. 80%. The method has been successfullyapplied to the assay of Shenfu injection (acompound preparation of Panax ginsengroot and aconite root). The recovery ofaconitine and aconite alkaloids is 95.68±1. 84-97. 78±1. 01%. The procedure is as follows: 10.0 ml ofShenfu injection was pipetted into a sepa-rator. The solution was rendered to pH 10-11with ammonium hydroxide and aconitealkaloids was extracted with five portions ofchloroform. The combined extract wasevaporated to dryness. The residue wasdissolved with a few drops of hydrochloricacid, and then was diluted to 10 ml withwater in a volumetric flask. For determination, to 2.0 ml of thesample solution in a separatory funnel wereadded 1.0 ml of distilled water, 5.0 ml ofphthalate buffer solution (pH 6.0), 2.0 mlof bromothymol blue indicator solution and10.0 ml of chloroform. The mixture wasshaken for 3 min and allowed to stand for1h. The chloroform layer was separatedand filtered. The absorbance of the chloro-form solution was measured at 410 nm,against a reagent blank. The content wascalculated from a standard curve preparedin the same manner with amounts ofaconitine.

The investigation shows that,with the addition of 4-8 drops of the mixed indicator,when the end- point is obtained,the pink color of the aqueous phase becomes blue and the blue color of the chloroformic phase becomes green.The both color change can be observed clearly.The two- phase titration has many advantages such as obvious end- point,low disturbance,easy- to- operate and good repeatability.In addition,its relative standard deviation is less than 1.0%.

Objective:To establish a method for determination of free sulfur dioxide and total sulfur dioxide in wine by automatic potentiometric titration.Methods:The wine samples were titrated without distillation operation.The sudden change of potentiometry in place of starch indicator could be judged at the end point of titration.Results:The RSD was 0.53%~0.76%,the recovery rate for free sulfur dioxide was 90.4%~99.4%.Compared with standard method,the relative deviation for total sulfur dioxide was less than 1.0%.Conclusion:The method is simple,accurate and without distillation operation.It is suitable for direct titration of the colored solution,the muddy solution and the inappropriate indicator solution.

目的:建立自动电位滴定法直接测定葡萄酒中游离SO2、总SO2的方法。方法:采用直接取样滴定,电位的突跃取代了淀粉指示剂来判定滴定等当点,测定出葡萄酒中游离SO2、总SO2含量。结果:该方法的变异系数为0.53%~0.76%,游离SO2测定回收率为90.4%~99.4%,总SO2测定与国标法相对相差不超过1.0%。结论:自动电位滴定法无需蒸馏处理,滴定简单准确,自动化性能高,对有色溶液、浑浊以及没有适合指示剂的溶液均可直接滴定。
