A new enzyme immunoassay system, Urease-antibody conjugate ELISA and its application in the diagnosis of schistosomiasis japonica was reported. A new enzyme immunoassay system, Urease-antibody conjugate ELISA and its application in the diagnosis of schistosomiasis japonica was reported. Urease was extracted and crystallized from jack bean meal. Urease-conjugate was prepared by the one-step glutaral-dehyde method. The substrate solution was made up with bromocresol purple and urea. Urease catalyzes the hydrolysis of urea to NH3, and pH shift causes a vivid colour change from yellowish to purple. This enzyme-substrate system offers a number of advantages, the substrate is stable, easy to obtain and without harm to human beings, and the colour change between negative and positive reaction is unmistakable. As urease is unaffected by the presence of the preservative sodium azide in reagents, the substrate may be prepared and stored until use. As urease does not exist in mammalian tissues, no endoge-neous activity occurs when urease-antibody conjugates are used to detect antibodies. A total of 36 normal persons and 36 schistosomiasis patients were tested by ELISA using urease-conjugate, the preliminary results showed that both sensitivity and specificity are promising.

By complex mutagenesis of Asp. usamii B_1 with N(?)G, UV—raysand LiCl, a black-spore mutant strain, B_(1-12), was selected. The strain is Suitable for liquid culturatation with a shorter fermentation circle and optimum pH shift to a lower pH value. The activity of exo-PG increases while the activity of glucoamylase decreases. But the stability of pectic enzymes at 60℃ decreases. The mutant strain is superior to B_1 in clarifying several kinds of fruit juices.

B_(1—12)是由Asp·usamii B_1经NTG、UV、LiCl复合处理选育到的一株呈黑色的孢子是适于液体培养的高酶活果胶酶菌株。具有发酵周期短、易过滤等痔点,酒樁沉淀的酶制剂与B_1比较,酶作用的最适pH分别为4.0和4.5,最适温度均力50℃,但在30℃时,B_1仍保持较高酶活水平的稳定性。B_(1—12)果胶酶组分ex—PG酶活高于B_1 0.5—1倍左右,endo—PG和PE变化不明显.两者都含有糖化酶、纤维素酶、蛋白酶、花青素酶等非果胶酶组分,其中糖化酶活性比B_1下降一倍.应用于苹果醋、苹果汁、草莓汁、樱桃汁的澄清.B_(1—12)优于B_1,透光率可达90%以上,但对枣汁的澄清效果却次于B_1。

The pitting corrosion resistence of different sensitized 304 stainless steels was investigated by means of electrochemical technology. The effect of NaCl concentration, temperature and pH on pitting potential (E b) was discussed. The results indicate that sensitization leads to E b decrease. Increasing NaCl concentration and temperature or decreasing pH shifts E b to more negative values. The intergranular corrosion of stainless steel makes pitting corrosion resistence worse.

用电化学测试技术研究了不同敏化处理的304不锈钢的耐孔蚀性能,探讨了Cl-浓度、温度和pH值对孔蚀电位Eb的影响。结果表明,敏化热处理导致Eb值下降;随Cl-浓度增加,介质温度升高,pH下降,使Eb值变负;不锈钢的晶间腐蚀使耐孔蚀性能变劣
