A stochastic analysis has been made of enzyme-substrate combination reactionsystems.The relationship between diffusion-encounter and effective collision inliquid phase fast reaction systems was discussed.A new formula,derived for calcula-ting the combination rates of multiple-active-site reaction systems,may serve toprovide the theoretical basis for experimentally investigating the combination activa-tion energies of liquid phase fast reaction systems.The results calculated for the fumarase-fumarate reaction system according to theformula are as follows:When the combination activation energy is small,the second-order rate constant calculated under the situation when the four active sites are allopen are nearly the same as that when only one active site is open;only when theactivation energy is large enough will the former be four times the size of the latter.Such a conclusion may also be extended to cover general multiple-active-site reactionsystems.This means that the usual method employed to estimate the combinationrates of multiple-active-site reaction systems is applicable only when the activationenergy is large,but not so when small.In this latter case,the coupling effect amongactive sites cannot be overlooked.Moreover,the transient phase process of enzyme-substrate reaction system hasalso been discussed briefly.

The active-site-carboxymethylated glyceraldehyde-3-phosphate dehydrogenase(GAPDH)from snake muscle when irradiated with ultraviolet light in the presenceof NAD~+ gives rise to a fluorescent derivative.The corrected emission maximum is420 nm and the corrected excitation spectrum shows two peaks at 240 nm and 285nm and a shoulder at 325 nm.The optimum conditions for the formation of the fluorescent derivative are aratio of 60 between the concentration of NAD~+ and CM-apo-GAPDH and irradiationat pH 7.6~8.0 for 8 minutes.A plot of the fluorescence intensities against theNAD~+ concentration shows a break in the curve at 4 moles of NAD~+ per moleenzyme.By fluorescence titration it is shown that binding of NAD~+ to snake muscleCM-GAPDH exhibits much weaker negative cooperativity than snake muscleGAPDH.A_(280)/A_(260) ratio of the snake muscle GAPDH fluorescent derivative is 1.61~1.63,which indicates that the irradiated snake muscle derivative contains two mole-cules NAD~+ per enzyme tetramer.The formation of the snake muscle GAPDH fluor-escent derivative is a“half-of-the-sitereaction similar to that of rabbit muscleGAPDH.

As in the case previously reported with the rabbit muscle enzyme, the carb oxymethylated lobster muscle glyceraldehyde-3- phosphate dehydrogenase also forms a similar fluorescent NAD derivative when irradiated with ultraviolet light in the presence of NAD+. The effects of pH on the formation, fluorescent intensity and stability of this fluorescent NAD derivative have been studied. In citr ate-phosphate-borate buffer, the formation, fluorescent intensity and stability of this fluorescent NAD derivative have pH optima at 7.0, 5.5 and 6.2 respectively whereas in phosphate alone, pH has very little effect on its formation, fluorescent intensity and stability within the range of pH 5-7. It appears that in aci dir solutions citrate inhibits the formation of this fluorescent NAD derivative, quenches its fluorescence emission and decreases its stability. The stoichiometry for the formation of the fluorescent derivative and the subsequent decarboxylation have been studied by phosphate analysis as well as by following the decarboxylation with the 14C-l-iodoacetate carboxymethylated enzyme. Like the rabbit muscle enzyme, both reactions are "half-of-the-sites" reactions. Unlike the rabbit muscle enzyme crystallized at pH 8.3 during the entire course of preparation of the lobster muscle enzyme, the pH has been kept below 7. This excludes the possibility that the stoichiometry for the formation of this fluorescent NAD derivative is an artifact arising from minor injuries to the enzyme molecule during crystallization at an alkaline pH.
