Using PAGE method, we analysed genomic RNA patterns of several standard strains of rotavirus including Wa, Ito, Yo Hochi, ST, SA-11 and UK strains, and the epidemic strains of rotavirus which caused infant diarrhoea in Nanjing in October to December 1989. It was suggested that the mutations and rearrangements of the genome of Wa strain may occur The different standard strains of human rotavirus(Ito, Yo, Hochi strain)and bovine rotavirus(UK strain) showed the same genomic RNA patterns, but the same standard strains(Wa, SA-11strain)obtained from different laboratories expressed different genomic RNA patterns. Besides, we proved that the human rotavirus was the major pathogen of the infant dirrhoea in Nanjing in the winter in 1989. The positive rate was 45.1%. The long electropherotype of human rotavirus was the dominant epidemic strain(89.2% ).

用聚丙烯酰胺凝胶电泳法对轮状病毒(RV)标准株进行RNA基因图谱分析,发现人轮状病毒(HRV)Wa株在MA104细胞上连续传5代后,其RNA基因图谱由原来的4:2:3:2模式变为4:3:3:2模式。继续分别在CV-1、MA104细胞上传代后,基因图谱进一步演变为4:3:4:2模式。经比较电泳、病毒空斑纯化,初步证实RVWa有变异林存在。目前尚未见RV标准株变异的报告,有待进一步研究。同时对多个RV标准株及1939年南京市区婴幼儿秋季腹泻的RV流行株进行基因图谱分析,发现不同的人RV及牛RV标准株有相同的基因图谱;不同实验室来源的同一标准株有不同的基因图谱。此外,尚证实HRV是1989年南京市区婴幼儿秋季腹泻的主要病源,总检出率45.1%,电泳长型为流行优势株89.2%。本文尚对RV RNA基因图谱变异的原因及意义进行了讨论。

The difficulties of rotavirus(RV)propagation in cell culture are not overcome completely as yet.For this reason screening a new cell line for RV propagation was carried out.After RVs were inoculated in the cultures of Frhk-4 cells,distinct cytopathogenic effects(CPE)developed.RV antigens and nucleic acids were detected in the infected cultures by ELISA and PACE,respective- ly.RV particles were observed on the ultra—thin sections of the infected Frhk—4 cells by EM. The results indicate that Frhk—4 is a new cell line susceptible to RV and may be a useful model for study of RV infection.In addition,the effects of concanavalin A(Con A)on RV propagation were observed under different conditions.The plaque number,productive titer and CPE degree of Wa strain significantly increased when certain dosages of Con A were added into the cell culture medium to pretreat the cells in the process of growth.The plaque number,however,significantly decreased under other conditions.Thus Con A is suggested to enhance or inhibit the RV multipli- cation in vitro under different condition.Moreover,it is interesting that the electrophoretic model of the genomic RNA of Wa strain changes after being cultured on MA104 cells for several pas- sages.Three variants were cloned from the“Wa”strain with the electrophoretic model of 4:3:4: 2.The genomic RNA of the variants was composed of 11 RNA segments,but the electrophoretic patterns were different from each other and that of Wa strain.The results suggest that the muta- tions and rearrangements of the genome of Wa strain may occur in vitro.

轮状病毒(RV)细胞培养问题迄今尚未完全解决。为此,我们从筛选RV 敏感细胞和改变 RV 培养条件入手研究,发现恒河猴胚肾传代细胞 Frhk-4对不同血清型的人和动物 RV 标准株敏感,感染后有明显 CPE。将刀豆素 A(Con A)加入营养液中,在细胞生长过程中预处理细胞时,能显著增加 Wa 株的空斑数目和增殖滴度,加速 CPE 的发展,而用 Con A 预处理病毒、加入病毒接种液或加入凝胶覆盖物时,则明显抑制 Wa 株的空斑形成。此外,我们发现,Wa 株经细胞培养多次传代后,有变异株产生,所分离到的3个克隆株的基因图谱与原始 Wa 株有明显差异。对此,国内外均未见报道。
