The polyethylene glycol (PEG) beaded phase has been employed for biologically active proteins by high performance hydrophobic interaction chromatography. The polyethylene glycol (PEG) beaded phase has been employed for biologically active proteins by high performance hydrophobic interaction chromatography. In this paper, the effects of Pore size of the silica and the PEG ligands with different molecular weight on coverage of the bonded phases and the resolution of protein separation were investigated. The experimental results illustrated that the PEG bonded phase with larger pore size gave higher coverage and better resolution- Moreover, comparison of resolution of the proteins tested in this work reveals that the bonded phase with smaller PEG ligands is suitable for the separation of hydrophobic proteins. In contrast, with larger molecular weight PEG ligand, the better resolution can be obtained. PEG-1500 coupled on the 50nm Lichrospher (10μm) gives the best resolution for the Protein separation. The activity recovery of the protein that obtained on this column exceeds 90%. 2mg protein sample can be injected into a PEG-1500 column (100μ5mm I. D.) without significant changes in resolution and retention time.
