A rapid liquid phase enzyme-linked immunosorbent assay (ELISA) method has been established for the identification of Xanthomonas campestris pv. oryzicola ( x. c. pv. oryzicola ) , and successfully used in the identification of naturally infected rice seeds. A rapid liquid phase enzyme-linked immunosorbent assay (ELISA) method has been established for the identification of Xanthomonas campestris pv. oryzicola ( x. c. pv. oryzicola ) , and successfully used in the identification of naturally infected rice seeds. In this method, x. c. pv. oryzicola (mixture bacteria) isolated from infected rice leaves in the fields of three provinces was used as the antigen (Ag) to immunize rabbits. The antiserum against x. c. pv. oryzicola was produced in the rabbits after eight intravenous injections during two months, with antiserum dilution as high as 1:9000. Horserodish peroxidase (HRP) was used to label protein A by the periodate sodium method. The pellets of Ag-Ab-HRP-protein A were precipitated by centrifugation. The process of determination for a batch of samples could be finished in three hours by this method. The results showed that this ELISA method had high specificity against x. c. pv. oryzicola, and hardly had crossreaction with nine other pathogenic bacteria of rice and other crops. The sensitivity of the method was approximately 103 cfu/ ml. The intrabatch and interbatch coefficients of variation were less than or equal to 15. 2% and 24. 5% respectively. Of the 363 seed samples collected in the infected fields of five provinces in South China, 297 were positive, representing a total positive rate of 81. B%. In Jiangxi province the rate of positive reaction on bacterial leaf stripe diseased rice plants was 92. 8%, while on symptomless rice plants in disease spread field was 63. 2%. This method has proved sensitive, specific and reliable, and is expected to be developed into a commercial ELISA kit for identification of infected rice seeds, investigation of the infection and spreading ways and early prediction of the disease.

介绍一种水稻细菌性条斑病病原菌的液相酶联法,可有效地应用于自然稻病种的鉴定。采用海南、浙江及江西三省稻区细菌性条斑病混合菌株为抗原免疫家兔,获得效价为9000倍的抗血清。该抗血清特异性强,对常见十余种植物病原细菌基本上无交叉反应;以酶标A蛋白为关键试剂建立的液相酶联法的灵敏度约为10~3cfu/ml;样品检测批间和批内变异系数分别小于24.5%和15.2%。该方法对部分省市419份自然病种的阳性检出率为83.3%;与目测田间发病的符合率达92.8%,而与目测田间未发病的符合率达63.2%。除江苏省外,来自南方四省的363份病种中,297份为阳性。该方法可识别因热处理、紫外线和γ射线以及三氯异氰尿酸等药剂处理后的死亡病菌,是一种简便、快速、灵敏、特异的可靠检测方法,可望制成商品诊断试剂药盒普及推广。

A rapid mini-preparation method for RNA isolation (MRRI) by acid guanidinium thiocyanatc-phenol-chloroform-liquid nitrogen-extraction is described. The DNA-RNA combining method for DNA and RNA isolation (DRCI) in uniform cells by acid guanidinium thiocyanate-phenol-chloroform-liquid nitrogen extraction was adapted. We also adapted a rapid liquid hybridization method (RLH), a new rapid procedure that DNA or RNA hybridized to ~(32)p-labeled probe after denature annealing-renaturation of sample and probe in Eppendorf tube, followed by 1.0% agarose gel electrophoresis, agarose gel dryer and autoradiography. The expression of GST π and three oncogenes in P388/ADR resistant lines by using the MRRI、DRCI and RLH method had showen that the expression of GST π RNA in P388/ADR resistant lines was increased by 1.56-fold (p<0.05), whereas the amplification of c-myc, v-erbB, N-ras oncogenes was decreased by 1.33, 1.38, 1.60-fold(p<0.05) respectively as compared with P388/0 sensitive lines.

本文采用一种小量快速RNA制备法(硫氰酸胍-酚,氯仿/耳戊醇-液氮)和DNA-RNA联合提取法(硫氰酸胍-酚,氯仿/异戊醇-液氮法),提取RNA和同一样本中DNA与RNA,同时采用一种快速液相杂交法,通过标记探针与样品DNA或RNA经变性-复性杂交、凝胶电泳、凝胶抽干、放射自显影。经用本法研究P388/ADR耐药株中的GSTπ及三种癌基因表达表明:耐药株中的GSTπ较敏感株增加1.56倍(p<0.05),而c-myc、v-erb-B、N-ras癌基因的扩增分别减少1.33,1.38和1.69倍(P<0.05)。
