The present paper is dealing with a rapid method of micronuclei test for testing chromosomal damage.This method was used for testing the time-effect and dose-effect of the Co60 r-irradiation on the frequence of micronuclei of the lymphocytes in rat blood.And the late effect of 265 rads Co60 r-irradiation in dogs was observed.The results showed that the effect of the radiation on the frequence of micronuclei of lymphocytes in blood and nucleated cell of bone marrow was consistent with the effect on chromosome...

本文介绍了用明胶-离心法分离和浓缩血液中淋巴细胞而后制片观察分析其微核出现率,并以此来测定染色体损伤的新方法。应用这种方法测定了大鼠受Co~(60)-r射线照射的剂量-效应时间-效应;还对受265拉特照射后活存狗的远期效应进行观察。结果表明,与骨髓有核细胞微核测定以及染色体畸变分析的结果是一致的。本文还对其应用价值作了初步的讨论。

用光镜和电镜技术研究了HL-60细胞在诱导分化过程中的显微与亚显微结构变化,10~(-6)M的维A酸处理6天,细胞按粒系途径定向分化,其核质比例降低,核浓缩、分叶,核仁减少或消失。经RA处理的细胞在电镜下出现下列明显的变化:细胞核浓缩和分叶,异染色质区域增加,约46%细胞显示出类似成熟粒细胞核的亚显微形态特征,胞质中嗜天青颗粒减少,特异颗粒显著增加,两种颗粒的比率发生明显变化;细胞质中微管、微丝的量增加;多聚和单个分散的游离核糖体减少,有些??细胞胞质空泡化;出现主要以微丝为筑架的大型钝形伪足和不规则的表面突起。上述这些变化似可作为HL-60细胞形态分化的标志。维A酸诱导HL-60细胞形态分化具有明显的时间效应关系。1.4%DMSO对HL-60细胞分化的诱导作用类似于10~(-6)MRA,而等剂量的(10~(-6)M)R Ⅰ、RⅡ其作用弱于RA。

The experimental results showed that toad nuclei aberration rate existed a significant linear dose-response relationship in certain region. The results of data on time-effect showed that the nuclei aberration rate reached maximum 12 hr. after the toxic chemicals were injected.In addition,by comparison of nuclei aberration rate with micronuclei, frequency the author suggested that the nuclei aberration [test was more sentitive and more reliable screening system than micronuclei test in monitoring environmental mutagenes and ecotoxicity.Nuclei aberration test will probably become a new to-xicity index in the toxicity test of environmental pollutants, and a new research method for amphibian ecotoxicology.

本文研究了大蟾蜍(Bufo bufo gargarizans)一次性腹腔注射环磷酰胺,甲醛周血有核红细胞畸核试验的剂量—效应时间效应。结果表明,在一定范围内,畸核率与药物浓度存在着明显的线性剂量—效应关系。时间效应实验数据显示,化学毒物注射后12h,畸核率达到高峰。畸核率与微核率比较,作者认为以畸核试验作为一个监测系统来监测环境诱变物和生态毒物比微核试验更敏感,更可靠,可望能作为环境污染物毒性测试的一个新的毒性指标,并为两栖类生态毒理学研究提供一个新的研究方法。
