The primary culture of rat Sertoli cells was used in this study to assay the toxicity of diethylstibestrol,endosulfan and cadmium chloride on rat Sertoli cells.Sertoli cells were isolated from testes of 28-day-old rats and cultured in the presence of various conventrations of these toxicants for 24,48,96hr.We found that diethylstibestrol,endosulfan and cadmium chloride decreasing the survival rate of Sertoli cells with relevant dose and time-effect relationship.The study also found that the viability of Sertoli cells from cadmium chloride-exposed rats was significantly decreased (P<0.05).This study supported that Sertoli cells could be the target cell of cadmium chloride in agreement with the study in vivo.This study showed that the primary culture of Sertoli cells was a rather stable and sensitive method for the investigation of potential testicular toxicants.It may be used in the screening of testicular toxicants and the study of toxic mechanism.

本研究在建立大鼠睾丸支持细胞体外培养方法的基础上,观察了不同浓度的硫丹、氯化镉对睾丸支持细胞的毒作用,发现它们对大鼠睾丸支持细胞均有毒作用,且有良好的时间效应和剂量效应关系。另外还研究了皮下注射20μmol/kg的氯化镉,24h后对所分离培养支持细胞存活的影响,发现体内给予氯化镉后,大鼠的支持细胞在体外培养4h后存活率明显低于对照组(P<0.05)。随着培养时间的延长,其存活率差距逐渐增大,说明氯化镉可以透过血睾屏障到达支持细胞。本研究表明大鼠睾丸支持细胞体外测试系统具有快速、简便、终点明确、良好体内外一致性等优点,可用于雄性生殖毒物的筛试和毒作用机理的评价。

Objective The aim of this study was to investigate the influence of lycopene on human prostatic cancer cell line DU145.Methods Time and dose effects of lycopene were examined by using trypan blue exclusion staining;The influence on the cell cycle and the apoptosis were observed with flow cytometry.The effect of oral intake of lycopen on the growth of DU145 tumors in nude mice was observed.Results Lycopene could inhibit the growth of DU145 cell line with a dose\|effect and time\|effect relationship.The rate of cell growth inhibition was 10\^76%,92\^9% and 99\^11%( P <0\^01) in those cells treated with lycopene for 7 days.In vivo the significantly inhibitied growth( P <0\^05)was observed on the male BALB/C nude mice having DU145 tumors.The rate of inhibition was up to 75\^76% ( 300 mg/kg ).The lycopene\|treated cancer cells failed to form tumors in nude mice.Lycopene can influence the cell cycle of DU145 and induce apoptosis.Conclusion Lycopene inhibits DU145 cell line independent of androgen in \%vitro\% and in \%vivo\%,and the mechanisms may be related to its influence on the cell cycle and inducing of apoptosis.

目的 研究番茄红素对人前列腺癌细胞DU145的影响。方法 体外培养的人前列腺癌细胞DU145经番茄红素作用后用苔盼蓝染色进行活细胞计数 ,绘制生长曲线并计算抑制率 ;应用流式细胞仪分析番茄红素对DU145细胞周期和凋亡的影响。制作裸鼠人前列腺癌模型 ,观察番茄红素在整体动物上的作用。结果 番茄红素可以明显抑制DU145细胞的生长 ,且呈剂量 效应时间 效应关系。 10、2 0、40 μmol L处理细胞 7d后 ,细胞增长的抑制率分别为 10 76 %、92 90 %、99 11% (P <0 0 1)。裸鼠人前列腺癌模型上的研究显示 ,6 %番茄红素油树脂可显著抑制腺瘤的生长 ,30 0mg kg剂量组腺癌抑制率可达到 75 76 % (P <0 0 5 )。用经番茄红素处理过的细胞致瘤力消失。流式细胞仪分析显示番茄红素可影响该细胞周期并引起细胞凋亡 ,凋亡率高达 42 42 %。结论 番茄红素可以在离体细胞和整体动物水平明显抑制雄激素非依赖型人前列腺癌DU 145 ,其抑制机制可能是通过影响人前列腺癌细胞的生长周期和诱导其凋亡而实现。
