Based on the high efficienct regeneration system of tomato, 3 kinds of screening reagents-kanamycin(Kan), neomycin(Neo), and herbicide PPT were respectively added into each medium of tomato tissue culture through different massconcentrations during the stages of the callus formation of tomato's cotyledon and hypocotyl, the differentiation and growth of tomato's bud, and the rootgrowth of tomato plantlets, so that the optimum screening massconcentrations needed by genetic transformation of tomato could be determined. The experiment showed that there was not distinct difference in resistance to above reagents between the two selfbred lines-T79 and T151; at each stage of tomato tissue culture, both kanamycin and PPT had a very stronger inhibition on explants, while neomycin had no obvious inhibition on them; on the other hand, during the different stages, there existed obvious difference in different explants' reaction to kanamycin and PPT, and particularly the callus formation of hypocotyl was the most susceptible to Kan, though the callus formation of cotyledon and the rootgrowth of plantlets were unsusceptible to it; in addition, the callus formation of hypocotyl and the differentiation of buds were susceptible to PPT, though the callus formation of cotyledon was unsusceptible to it. Hence, it was suggested that for genetic transformation of tomato, Kan 100 mg·L-1 should be selected as ideal screening massconcentration at the stage of hypocotyl callus inducing and plantlets' rootgrowth, Kan 50 mg·L-1 at the stage of hypocotyl callus inducing; and Kan 70 mg·L-1 at the stage of bud differentiation. However, for PPT, 1.0 mg·L-1 should be taken as ideal screening massconcentration at the stage of cotyledon callus inducing, 0.5 mg·L-1 at the stage of hypocotyl callus inducing; and 0.75 mg·L-1 at the stage of bud differentiation and plantlets' rootgrowth.

Objective- To investigate the efficacy of antihypercoagulability state enzyme (AHCSE) on human hepatocellular carcinoma BEL-7404 cells and probe into its function. Methods- MTT. Phase-contrast microscope, electron microscope, terminal deoxynudeotidy transferase dUTP nick and labeling (TUNEL) were conducted to observe the alteration of cell form and biochemistry variation of the AHCSEed BEL-7404 cells. Results The AHCSEed BEL-7404 cells altered in its forms. The low dosage AHCSE had a very strong inhibition to BEL-7404 cells, but with the dosage increase, the rate of inhibition did not rise markedly. After the action of AHCSE, Apoptosis took place in BEL-7404 cells, the apoptosis index rose with dosage increase. Conclusion- The AHCSE has very strong inhibition on the BEL-7404 and inducing apoptosis is one of its functions.

目的研究蛇毒抗高凝状态酶(AHCSE)对人肝癌细胞BEL-7404的作用及其可能的作用机制。方法应用相差显微镜、电子显微镜、MTT法、TUNEL法等方法观察BEL-7404细胞经过AHCSE处理后,细胞形态学、生物化学等方面的变化。结果人肝癌细胞BEL-7404经过AHCSE处理后,发生了形态学改变,胞浆内部分线粒体空泡化,成簇,胞膜下多见。浓度为0.5μg/ml的AHCSE对BEL-7404细胞即有很强的抑制作用,随着剂量的加大,其抑制率上升不明显。经AHCSE作用后,BEL-7404细胞发生了凋亡,凋亡率随AHCSE浓度的增加而增加。结论AHCSE对BEL-7404细胞具有很强的抑制作用,诱导细胞凋亡是其作用机制之一。

OBJECTIVE:To research the effect of antihypercoagulability state enzyme (AHCSE) on human hepatocellular carcinoma BEL-7404 cells and SMMC-7721 cells, normal Hepalocellular LO2 cells and probe into its function. METHODS: MTT, phase-contrast-microscope, electron-microcopy, terminal deoxynudeotidy transferase dUTP nick and labeling (TUNEL) and flow cytometer (FCM) were conducted to observe the alteration of cell form and biochemistry variety of the AHCSEed cells of BEL-7404 cells, SMMC-7721 cells and LO2 cells. RESULTS: The AHCSEed cells of BEL-7404, SMMC-7721 altered in their cells forms. The suspending cells and the cellular debris got more, the process of cell division got less. Part of mitochondrion became hollows and clusters. There were myeline sheath-liked structures in cytoplasm. The 0.5 μg/mL dosage AHCSE had a very strong inhibition to BEL-7404 cells (P=0.000). SMMC-7721 cells (P=0.025), but nearly no efficacy on LO2 cells (P=1.000). With the dosage increased, the dosage AHCSE was 75 μg/mL, the rate of inhibition to BEL-7404 cells and SMMC-7721 cells did not rise significantly, P=0.095, 0.115, but an inhibition to LO2 cells appeared (P=0.049). After the action of AHCSE apoptosis took place in BEL-7404 cells (P=1.000) and SMMC-7721 cells (P=1.000), and the apoptosis index increased with the increase of the dosage (P=1.000) and time (P=1.000). CONCLUSIONS: The AHCSE has very strong inhibition to the BEL-7404 cells and SMMC-7721 cells, but little efficacy on LO2 cells. The efficacy of AHCSE on hepatocarcinoma is safe and effective. Besides the direct damaging effect, inducing apoptosis is one of its functions.
