IM: To build a structure model for the pore of voltage gated Shaker potassium channel and examine its validity. METHODS: (1) Structural restraints were derived from experimental and theoretical studies; (2) An initial structural motif satisfying the derived restraints was first constructed, and further refined by restrained molecular mechanics; (3) The quality of the model was judged by the criterion that whether it could clarify molecular mechanisms of channel functions and explain the known experimental facts. RESULTS: (1) A computer pore structure was proposed, in which the residues within signature sequence (corresponding to Shaker 439-446) dipped into the membrane and formed the narrow part of the pore in a non periodic conformation, while the other residues in the P region constituted the outer mouth of the pore; (2) The ion selectivity was achieved through cation π orbital interaction mechanism at position 445 and oxygen cage mechanism at position 447; (3) Different binding modes led to different affinity of CTX and AgTx2 to channel; and (4) The inside of pore was dominated by negative electrostatic potential. CONCLU ̄SION: The model proposed was consistent with the derived restraints from the experimental results.

In order to search the effective medicine to delay the advance of chronic renal failure, the paper reported the observation results on applying yishen fuyuande-coction which had the warming kidney,. replenishing essence, promoting and toni-fying extra-channels function to treat patients (22 cases)with chronic renal failure, meanwhile took western medicine and western and Chinese medicine as the-controll groups. The results showed that the advance was linear relation ship tol/scr, the levels of 1/scr of...

The results showed that the advance was linear relation ship tol/scr, the levels of 1/scr of three groups lowed slowly, this suggested that the three therapy could delay the advance of chronic renal failure. Regression e-quation analysis showed that slope (b) of western and Chinese medicinegroup was the biggest among three groups. All theses indicated that yishenfuyuan decoction could delay the advance of CRF,the curative effect of decoction and western medicine was the best. The decoction could increase the levelsof patients' Hb., ALb, Ca2+, decrease BuN>P3",this suggested the decoction could im-provethe survival condition of patients with CRF.

为了寻找延缓慢性肾功能衰竭进展过程的有效药物,运用温肾填精、通补奇经中药益肾复元汤治疗CRF患者22例,同时以巯甲丙脯酸片及中西药合用做对照。结果表明与CRF进展呈线性相关的1/Scr,三组病人在治疗1、3、5、7、9、11个月后与疗前比较下降缓慢(P>0.05),提示三种治疗均有延缓CRF进展作用。回归方程分析显示三组斜率b值中西药组最大,中药组次之,西药组较小(P<0.05),由此证实益肾复元汤确有延缓CRF进展的作用,与西药合用,效果更佳。并且治疗后中西药及中药两组Hb、ALb、Ca~(2+)水平较疗前均有提高,BuN、P3~-均有降低,西药组无此作用,提示益肾复元汤还有改善CRF患者生存质量的作用。
