α-Fetoprotein (AFP) is a well known onco-fetal protein, which has widely been adoped in the diagnosis of liver cell cancer. Although the physico-chemical properties of both AFPs from fetuses and patients are identical, little information about thier biological functions is known up to date.In this paper, taking human and rat macrophages as materials, as well as chicken red blood cells as marker, we are going to report the effect of human AFP (HAFP) on immunological function.1. HAFP suppressed the phagocytosis of human macrophages and depressed its mobility while HSA did not show this potency.2. AFP positive sera from patients with liver cell cancer likewise displayed suppression of phagocytosis before AFP removal and this suppression can partially or totally be released after AFP removal.3. The potency of cells from rat peritoneal exudate to bind HAFP was much stronger than that to HSA; suppression of phagocytosis by HAFP was also much stronger than that by HSA.4. Binding ratio and binding amount curves showed: (1) When HAFP concontration was less than 1ng/200μl, the K_D is 2.5×10~(-12)M, and the interactions of HAFP with cells of rat peritoneal exudate (macrophages was 70~80%) might be positive cooparation. (2) When HAFP concentration was higher than 1ng/200μl some of HAFP molecules bound might be more relaxing.5. Peroxidase labelled immune electronmicrography showed that HAFP molecules were capable of distributing to the surface of macrophages.6. The effect that ~(131)I-HAFP could be displaced by HAFP showed the specificity of interaction of HAFP with macrophages.Based on the above findings it is suggested that HAFP receptors might exist on the surface of macrophages and result in the suppression of phagocytosis after binding with HAFP. It seems reasonable to postulate that HAFP is an immune regulating factor during the fetal development and in the normal adults while an immune suppressive factor in the patients with liver cell cancer.
