Allergic asthma caused by Pleurotus sapidus besidiospores occurs in high incidence and brings about great hazard among mushroom cultivating workers in China.In order to elucidate the role of cytokines in the pathogenesis of this disease,we set up an animal model of allergic asthma through the challenge of Pleurotus sapidus besidiospores in primed rats,and measured a series of related cytokines and their receptor mRNA levels of spleen lymphocytes from rats with asthma.SDS PAGE analysis showed that the besidiospore antigen is a 17kDa protein.Cytokines and their receptor expression at mRNA levels were determined by RNA/cDNA hybridization using radiolabelled cDNA probes for different cytokines and their receptors in vitro. Results showed that mRNA expression of IL 4,GM CSF,IL 6,IL 2 and IL 2R,but IL 6R in lymphocytes of asthma suffering rats challenged for 7d increased by 54%,45%,170%,83% and 76%,respectively.In rats challenged for 2d,mRNA levels of IL 4,IL 6 and IL 2 increased respectively by 37%,58% and 125%;whereas mRNA level of GM CSF,IL 2R and IL 6R unchanged.Results suggest that allergic asthma induced by Pleurotus sapidus besidiospores markedly activates IL 4,GM CSF,IL 6,IL 2 and IL 2R,except IL 6R expression in lymphocytes as evaluated at mRNA level.We consider that there exists a significantly increase in T H2 type cytokines in the pathogenesis of Pleurotus sapidus besidiospore induced asthma in which IL 4 may play an essential role.

提取平菇孢子抗原诱发动物过敏性哮喘,经ConA体外诱导脾淋巴细胞产生相关细胞因子,用同位素标记的各种细胞因子及其受体的cDNA作为探针进行RNA/cDNA分子杂交,研究哮喘大鼠脾细胞中细胞因子及其受体mRNA的表达水平。结果显示,激发7天的哮喘大鼠脾细胞中除IL-6R外,IL-4,GM-CSF,IL-6,IL-2和IL-2RmRNA的表达量分别提高了54%,45%,170%,83%和76%。而在激发2天的哮喘大鼠中,仅IL-4,IL-6和IL-2mRNA表达明显提高,分别为37%,58%和125%,但GM-CSF,IL-2R和IL-6RmRNA表达保持不变。此外,激发7天与激发2天的致敏哮喘大鼠相比,IL-4,GM-CSF和IL-6mRNA的表达量分别提高了13%,42%和71%,而IL-2mRNA的表达则下降了18%。说明从2天到7天的激发过程中,TH2细胞因子GM-CSF和IL-6mRNA提高的幅度远远大于IL-4,TH1细胞因子IL-2mRNA的表达反而下降。由此表明,平菇孢子致敏哮喘显著提高脾淋巴细胞IL-4,GM-CSF,IL-6,IL-2和IL-2RmRNA的表达。我们认为在平菇孢子致敏哮喘的病?

Thrombopoietin(Tpo) concentration-dependent proliferation of TD-3 celis of Mpl receptor gene transfected celi line was observed by means of MTT method for the purpose of establishing a new bioassay method to determine the biological activity of Tpo in samples or products in vitro. An optimized condition was obtained after comparison of various experimental conditions. The proliferation of TD-3 celis was highly selective with cy-tokines, only Tpo and mIL-3 promoted proliferation of the cells, but human IL-3 and other 7 megakaryocy-topoiesis related cytokines had no effect at all. The sensitivity of the assay was comparable to MO7e cell reported in the literature, the ED50 for Tpo was 1-3 ng/ml. The results indicated that TD-3 celi line could be used for bioassay of Tpo.

为建立体外检测血小板生成素(thrombopoietin,Tpo)生物活性的生物检定方法,采用MTT法观察了转染Mpl受体基因的TD-3细胞株依赖于Tpo浓度的增殖特性。通过观察实验条件对增殖的影响获得了最佳实验条件,比较了TD-3细胞对10种与巨核-血小板系造血相关的细胞因子的反应,表明该细胞株仅对Tpo和鼠白细胞介素3(mIL-3)有浓度依赖性的增殖反应,对其它细胞因子均无反应,表现出高度的选择性。检测Tpo的ED_(50)为1-3ng/ml,与文献报道检测Tpo生物活性的MO7e细胞株相近。以上特征表明该细胞株可作为检测Tpo生物活性的工具。

Conclusion In the mice infected by schistosome japonicum, the immune response of Th2 related cytokines took place mainly in the liver. The spleen played a role of immune regulation. As for colons, the increase of Th2 related cytokines may be only represented a part of immune reaction in whole body.

目的了解感染日本血吸虫后小鼠不同脏器Th2相关因子介导的免疫反应。方法采用ABC免疫组化法,并用多媒体病理图文定量分析,动态研究感染后IL4,IL5,IL10的变化。结果在感染小鼠肝脏IL4,IL5,IL10明显高于正常对照(P<0.01),并随感染时间的延长而逐步升高;在脾脏IL4,IL5,IL10亦升高,但其幅度明显较肝脏为低;在结肠IL4,IL5,IL10随感染时间延长呈缓慢升高趋势。结论感染日本血吸虫病的小鼠,肝脏是Th2相关因子免疫应答的主要器官;脾脏是调节免疫的器官;而结肠Th2相关因子的升高可能是整体反应的一部分
