    Hepatic 9000×g supernatant (S9) fractions derived from male Sprague-Dawley rats which had been pair-fed isocaloric ethanol or carbohydrate control diets for 4 weeks, were used to determine the effect of chronic ethanol feeding on hepatic inactivation of the mutage-nic activity associated with the primary carcinogen N-methy1-N′-nitro -N-nitrosoguanidine (MNNG). Ethanol feeding enhanced the capacity of hepatic S9 to inactivate MNNG mutagenicity as measured by Ames assay (p<0.05). The increased detoxificaion...

    Hepatic 9000×g supernatant (S9) fractions derived from male Sprague-Dawley rats which had been pair-fed isocaloric ethanol or carbohydrate control diets for 4 weeks, were used to determine the effect of chronic ethanol feeding on hepatic inactivation of the mutage-nic activity associated with the primary carcinogen N-methy1-N′-nitro -N-nitrosoguanidine (MNNG). Ethanol feeding enhanced the capacity of hepatic S9 to inactivate MNNG mutagenicity as measured by Ames assay (p<0.05). The increased detoxificaion of MNNG by rat liver S9 from ethanol fed rats was associated with increased activities of several detoxifying enzymes. The activities of GSH S-transferase and UDP glucuronosyltransferase in S9 from ethanol fed rats were 1.2 and1.7 times as in controls and the activity of a marker microsomal cyto-chrome P-450 dependent enzyme, aminopyrine N-demethylase, was 1.2 times as in controls. Ethanol feeding also produced an increase in hepatic GSH levels, but did not affect UDPGA levels. The present results suggest that the increased detoxifying capacity of the livers from ethanol fed rats for the direct acting carcinogen and mutagen MNNG is due to the increased activity of the above enzymes and the cofactor GSH.

    为了研究护肝宁口服液抗酒精中毒作用及其机理。采用雄性 SD 大鼠,每日饲饮10%乙醇8g/kg 体重,同时每日饲护肝宁口服液(生药40g/kg 体重)。结果表明,长期饮用乙醇的大鼠肝脏和睾丸组织 LPO 水平皆明显高于对照组(P<0.01),在饲饮乙醇之前给予护肝宁口服液的大鼠,肝脏和睾丸组织 LPO 水平不升高,明显低于乙醇组大鼠(P<0.01)。提示长期饮酒可以引起肝脏和睾丸组织过氧化损害,护肝宁口服液能有效地保护肝脏和睾丸免遭乙醇的损害。
