目的:探讨利用手指血取样代替静脉血对卡马西平进行治疗药物监测(TDM)的可能性。方法:应用反相高效液相色谱(RPHPLC)法。结果:患者静脉血与手指血卡马西平浓度二者相关性好(r=0.963 9),经统计学分析(配对t检验)二者差异没有显著性(P>0.05)。结论:应用HPLC法对卡马西平进行TDM,可以用手指血代替静脉血。

To develop a simple and rapid whole-blood immunoassay for detecting serum HBsAg,using an anti-HBsAg and anti-RBC bispecific diabody. 20 μl of finger prick, ear prick or heparinized blood was mixed with 50 μl bispecific diabody, and the autologous red blood cell(RBC) agglutination was monitored on slide or 96 well U shaped plate. Comparison of the two RBC agglutination methods showed that the slide assay was better than 96 well plate assay in simplicity and sensitivity, thus the former was used in the following experiments. The assay had a detection limit of at least 20 μg/L. Blood samples from 512 patients were tested using both auto-RBC agglutination assay and routine ELISA. No false positive results were found in 347 samples that were negative by ELISA. Among 165 ELISA positive samples, there were 4 false-negative(2.4%). The auto-RBC agglutination assay for HBsAg has an accordant rate of 97.6% with ELISA. Its advantages are speed and simplicity.

目的 用抗乙肝表面抗原 (HBsAg)和抗红细胞 (RBC)双特异Diabody ,建立一种简便、快速、用全血检测血中HBsAg的方法。方法 用病人指血 ,耳血 ,抗凝全血 ,或残留未凝血细胞的非抗凝全血 2 0 μl与 5 0 μl双特异Diabody上清混合 ,分别在玻片和 96孔U型板观察血细胞凝集。结果 实验的简便性和敏感性以玻片法为优。用凝集法与ELISA法平行检测病人5 12例 ,以ELISA法结果为标准 ,凝集法假阳性率 0 % (n =3 47) ,假阴性率 2 4 % ( 4 / 165 )。自身红细胞凝集法检测HBsAg与ELISA方法符合率 97 6%。结论 本方法的优点是快速、简便
