By immunogold technique the intracytoplasmic lysozyme content and distribution were analysed in 8 cases with acute promyelocytic leukemia (APL). The majority of granules in leukaemic cells lacked lysozyme activity, though hyperactive production of primary granules occurred. However, in some rather immature leukaemic cells were detected 1ysozyme-positive granules. In remission after the retinoic acid treatment intracytoplasmic granules exhibited frequently positivity for lysozyme with the maturation of leukaemic cells. It was confirmed that there existed abnormality of the granule components in APL cells besides overproduction of primary granules. The result suggested that the measurement of intracytoplasmic lysozyme might be helpful to judge the effect of induced differentiation therapy for APL.

本研究应用胶体金免疫电镜技术,观察了8例颗粒增多的急性早幼粒细胞白血病细胞内溶菌酶的分布及含量。观察中发现早幼粒白血病细胞中尽管有大量的初级颗粒生成,但这些颗粒多数缺乏溶菌酶活性,而少数形态较原始的白血病细胞中却查见溶菌酶阳性颗粒。维甲酸治疗后,细胞形态趋于成熟,细胞中颗粒常表现为溶菌酶阳性反应。本观察显示急性早幼粒细胞白血病不仅颗粒生成亢进,颗粒中组成成份也发生异常;并提示白血病细胞中溶菌酶活性的测定,对于判断诱导分化治疗白血病的疗效具有一定的参考价值。

An expression vector of this cDNA(pRA538)was co transfected with the neo gene(pDORneo)into parental cancer cell line EC8712.The cell colonies obtained after selection in G418-containing culture medium showed very poor growth,reduced(by 68%-76%)3H-TdR uptake and morphological changes characteristic of terminal differentiation and programmed cell death(apoptosis).In situ hybridization with RA538 probes revealed expression of mRNA of RA538 in the cytoplasm of the transfected cells.The cells transfected solely with pDORneo after G418 selection showed normal growth pattern and no RA538 expression. However,none of the control cells EC8712 survived the 6418 selection.Thus the expression of cDNA RA538 has a similar effect on the esophageal cancer cell EC8712 as the retinoic acid does.

Purpose: The PML-RAR α fusion gene resulted from the balanced t(15;17) translocation, as a molecular marker of acute promyelocytic leukemia, is postulated to be responsible for promyelocytic leukemo-genesis and all-trans retinoic acid treatment. The effects of Antisense oligodeoxynucleotides (ASODN) specialized to PML-RAR α and retinoic acid on acute promyelocytic leukemia cell line NB4 and HL-60 maybe show us something about the relation between the PML-RAR α fusion gene and ATRA treatment....

Purpose: The PML-RAR α fusion gene resulted from the balanced t(15;17) translocation, as a molecular marker of acute promyelocytic leukemia, is postulated to be responsible for promyelocytic leukemo-genesis and all-trans retinoic acid treatment. The effects of Antisense oligodeoxynucleotides (ASODN) specialized to PML-RAR α and retinoic acid on acute promyelocytic leukemia cell line NB4 and HL-60 maybe show us something about the relation between the PML-RAR α fusion gene and ATRA treatment. Methods: ASODN-mediated inactivation of the PML-RAR α mRNA was measured by RT-PCR. The proliferation and differentiation of NB4 cells were determined by proliferation curve, morphology, CD antigen of membrane and NBT test. NB4 Cell cycle was analyzed by FACS. Results: ①ASODN could only act as a specific growth inhibitor and partially differentiation-inducer to NB4 cells. ②The proliferation and differentiation of NB4 and HL-60 cells are both affected by retinoic acid. ③ASODN could decrease the percentage of S phase cell of NB4 cells, but retinoic acid couldn't. Conclusion: ASODN specialized to PML-RAR α and retinoic acid on the proliferation and differentiation of NB4 and HL-60 cell line may have different mechanism, and ASODN is more specialized and earlier than retinoic acid.

目的:急性早幼粒细胞白血病染色体t(15;17)易位形成的PML-RARα融合基因作为早幼粒细胞白血病的标志与APL的发病及维甲酸诱导分化效应密切相关。本研究将针对PML-RARα基因的反义核酸和维甲酸对NB4细胞和HL-60细胞生长分化的影响加以比较,探讨PML-RARα基因对维甲酸诱导分化作用的影响。方法:用反义核酸来封闭PML-RARα融合基因的表达并用RT-PCR的方法检测。NB4细胞的生长、分化及功能通过细胞生长曲线、形态学、细胞表面标志及NBT还原试验加以判定,细胞周期应用流式细胞仪进行分析。结果:①反义核酸仅能够抑制NB4的生长,促进NB4细胞的分化,具有特异性;②维甲酸能够同时影响NB4和HL-60细胞的生长分化;③反义核酸能够降低NB4细胞S期细胞百分比,而维甲酸不能。结论:ASODN和维甲酸影响APL细胞的生长分化作用机制可能不同,而且反义核酸作用特异性强,作用时相早。
