            应用Z-82A型抗生素抑菌圈面积测量仪,能精确测量薄层层析中生物显迹单组份的抑菌斑,并能计算多组份抗生素的组份百分率。以氨基糖苷类抗生素卡那霉素、新霉素和托普霉素为例,用烧结硅胶玻璃板和硅胶塑料膜进行薄层层析得到了满意结果。在测量多组份氨基糖苷类抗生素80-61号时,能正确计算出各组份的百分含量。本方法将为抗生素纯品检定和多组份抗生素的发酵、分离和纯化以及生物或化学转化工作提供准确的定量数据。在硅胶薄层层析中,点样的面积和点样的体积对展层后抑菌斑的面积实际影响不是太大。但因每块薄板的质量差异较大,故在每次实验中必须加一个标准浓度的对照,以便校正可能出现的误差,并尽可能采用一致的标准条件。

            从我国四川省九襄县土壤中分离到一株链霉菌83-1050B,经形态、培养特征、生理生化特性、拮抗性、全细胞及细胞壁化学组分等方面的研究表明,与文献报道的暗霉素产生菌——黑暗链霉菌Streptomyces tenebrarius ATCC 17920基本相似,认为是黑暗链霉菌的新株,定名为黑暗链霉菌83-1050B(Streptomyces tenebrarius 83-1050B)。该菌株产生多组分的氨基糖苷类抗生素复合物,具有广谱的抗菌活性,复合物的主要组分有三个,其中83-1050B-Ⅰ和83-1050B-Ⅱ经分离、纯化,测定理化性质及生物学活性,分别与文献报道的阿普拉霉素(Apramycin)和氨甲酰妥布拉霉素(6”-O-carbamoyl tobramycin)为同质。氨甲酰妥布拉霉素经碱水解得到妥布拉霉素,从体外抗菌活性结果说明:妥布拉霉素对各种细菌的作用比庆大霉素强,尤以抗绿脓杆菌的作用较突出,83-1050B-Ⅱ和庆大霉素作用相当,83-1050B-1的抗菌作用比庆大霉素差。
