We have developed previously aprocedure to detect the down-and up-regulation of NK cell activity by in vi-tro differentiated mature human macro-phages(Mφ)and LPS modulated Mφ.The aim of this study is to investigatethe dynamic changes of monocyte-Mφin the course of in vitro culture withrespect to their regulation of NK cellactivity and its possible mechanism. Human monocytes from peripheralblood mononuclear cells(PBMNC)ofhealthy adult donors were separated byplastic adherence as previously describedand cultured...

We have developed previously aprocedure to detect the down-and up-regulation of NK cell activity by in vi-tro differentiated mature human macro-phages(Mφ)and LPS modulated Mφ.The aim of this study is to investigatethe dynamic changes of monocyte-Mφin the course of in vitro culture withrespect to their regulation of NK cellactivity and its possible mechanism. Human monocytes from peripheralblood mononuclear cells(PBMNC)ofhealthy adult donors were separated byplastic adherence as previously describedand cultured first in RPMI-1640 mediumsupplemented with 20% AB serum for0,2,4,6 and 8 days separately.Theireffect on NK cell activity was thentested by mixing them with freshly iso-lated PBMNC and ~(50)Cr-labeled K 562tumor cells and measuring ~(51)Cr releasein a 4 hr short-term assay.The resultsshowed that with the increase of cellsize(Table 1),day 6 and day 8 macro-phages in culture were reproduciblyshown to have the property to down-regulate NK cell activity(more than50% suppression)as compared with day0,2 and 4 Mφs at a ratio of Mφ toPBMNC of 0.5:1(Table 2).This find-ing indicated that human blood mono-cyte-Mφ at different stages of differen-tiation/maturation may have a distinctfunction and that down-regulation of NKactivity by day 6 and day 8 Mφs canbe measured by a rapid and efficient 4 hrshort-term assay.The suppression of NKcell activity against K 562 cells by cul-tured Mφs was correlated with their levelof cytostasis toward P 388 tumor cells(Table 3). E-type prostaglandin(PGE)andoxygen radical productions(O_2~-,H_2O_2,OH)have been shown to be able toimpair NK activity,but in the presentexperiment,the presence of indomethacin(1×10~(-5)M),an inhibitor of PGE syn-thetase,and catalase(4000 units/ml),aH_2O_2 oxidoreductase,has not caused anylowering of the observablely impairedNK activity.This might mean that theobserved suppression of NK activity is ??not due to prostaglandins and/or freeoxygen radicals(Table 4).Furthermore,the failure to see any appreciable killingof ~(51)Cr-labeled day 6 Mφ by allogenicNK cells from normal individuals(Tab-le 5)tends to rule out the possibilitythat the suppression of Mφ against NKcell activity is HLA-restricted and thatMφs bear target structures which can berecognized by NK cells and competedwith those of K 562 tumor cells. Adherent monocyte-Mφ from day 0and day 6 were sonicated by FisherSonic Dismembrator Model 150.95—98%of viable Mφs was destroyed and sup-pression activity of sonicates was thenmeasured.No lowering,but even anincrease,of suppression of NK activityby day 6 Mφ sonicates was seen(Table6).Similarly,day 6 Mφ)sonicates alsoshown a strong cytostasis effect on P388 tumor cells(Table 7).These resultsindicated that differentiated mature hu-man Mφs may produce and release solu-ble mediator(s)to exert their action onNK activity and tumor cell cytostasis. The possibility of space barrier orartificial crowding effect induced by Mφscan also be ruled out by the fact thatmodulated day 6 Mφ by LPS actuallyenhanced the expression of NK activity(Table 8),because modulated and non- ??modulated day 6 Mφs are of same size.The above data again point out to thepossible release of enhancing factor(s)bymodulated Mφs and suppressing factor(s)by non-modulated ones.It may be ques-tioned whether all of the impaired NKactivity by mature Mφs in vitro can bemainly attributed to the release of solu-ble factors,but this appears to be highlypossible from the evidences at our own In sum,the results from this paperand from our previous works indicatedthat the regulatory function of Mφs onNK cell activity is correlated to theirstate of differentiation and the positiveand negative regulation of NK activitycan be attributed to different Mφ subpo-pulations. *1.5×10~5 PBMNC % NK activity:42.1±2.1.2.Treatment of macrophages by LPS(10μg/ml)for 18 hrs,then washing away LPS.Preincubation with PBMNC for 4 hr,measured ~(51)Cr CPM in supernatate.3.The experiment was repeated four times and the results of a representative experiment are givenhere.** % inhibition of NK activity is shown in parenthese.

On the basis of primary operation principle of near infrared vidi-con,improvement is made in target structure and techniques.The ex-perimental data show that the performances,especially the life time,of the improved near infrared vidicon are improved.

Twelve cases were treated by bilateral anterior cruna of the internal capsule as target structure for schizophrenia of effective disorder as depression, anxiety, obsession, etc. Eight cases were treated by using bilateral medial nuclei of the amygdala as the target structure for schizophrenia of excitation, impulsion and aggressive behavier as well as epileptic psychogeny....

报告20例精神分裂症的脑立体定向治疗。分为两组,以双侧内囊前肢为靶结构者12例;以双侧杏仁核的内侧核为靶结构者8例。术后随访1~2年,平均1.3年。显效7例;有效12例;无效1例。
